Objectives:This study aimed to investigate the influence of Quanzhenyiqi Decoction on chronic airway inflammation,lung pathology,and ultrastructural of COPD mice by observing the changes of ultrastructure and the expression of bronchoalveolar lavage fluid IL-6 and IL-8 while applying the Quanzhenyiqi Decoction to COPD mice.Method:The C57 mice were randomly divided into normal control group,model group,Quanzhenyiqi decoction low and high dose groups,and aminophylline group.COPD mice model was established by combining intratracheal instillation of LPS and exposure to cigarette smoking,which was lasting for 16 weeks.During the model establishing period,the general conditions of mice were closely observed,including the changes of pathology and ultrastructural of lung tissue.Besides,the total number of inflammatory cells in bronchoalveolar lavage fluid were counted,along with the measure of pulmonary function,expression of bronchoalveolar lavage fluid IL-6 and IL-8,and the ERK,Nrf2 and HO-1 protein in lung tissue.Result:1.General situations: Compared with the control group,the state of mice of model group was worse,and the body weight was reduced significantly,which has statistical sense(P<0.05).And compared with the model group,the state of mice of Quanzhenyiqi decoction low and high dose groups were become better with the body increased,especially the high dose group.2.Results of pulmonary function test: Compared with the control group,airway resistance of mice of model group increased,which has statistical sense(P<0.05).And the lung static compliance was significantly higher,which also has statistical sense(P<0.05).At the same time,Functional residual capacity(FRC)of model group increased,but didn?t have statistical sense(p>0.05).The change of airway resistance of mice of model group also has statistical sense(P<0.05),along with the decrease of lung static compliance and functional residual capacity.3.Lung pathological observation: In control group,the respiratory and alveolar structures of mice were normal and complete,without uncompensated emphysema and alveolar inflammation.However,in the model group,there were inflammatory cell infiltration around the bronchi,vascular dilatation and congestion,pulmonary interstitial hyperplasia,peripheral inflammatory cell infiltration,widening of alveolar septum,alveolar emphysema forming by rupture fusion of partial alveolar,and the compensatory emphysema formed.At the same time,compared with the model group,the lung tissue pathology of mice of Quanzhenyiqi decoction groups improved.4.The results of expression of ERK,Nrf2 and HO-1 protein in lung tissue: There was only a small amount of ERK protein expression in the control group;The expression of ERK protein in the model group increased significantly;The expression of ERK protein in the Quanzhenyiqi decoction low and high dose groups reduced significantly;There was only a small amount of Nrf2 protein expression in the control group;The expression of Nrf2 protein in the model group increased,while decreased in the Quanzhenyiqi decoction low and high dose groups;There was only a small amount of HO-1 protein expression in the control group and the model group,while increased in the Quanzhenyiqi decoction low and high dose groups.5.Results of total number of inflammatory cells in bronchoalveolar lavage fluid of mice and the expression of IL-6 and IL-8: Compared with the control group,the total number of cell increased significantly,and the concentrations of IL-6 and IL-8 increased,which have statistical sense(P<0.05).Compared with the model group,the total number of cell count decreased significantly and the concentrations of IL-6 and IL-8 increased in the Quanzhenyiqi decoction low and high dose groups,which have statistical sense(P<0.05).6.Results of lung tissue observed in electron microscope: The cell membrane,organelle and nuclear structure of mice remained normal and complete in the control group.In the model group,the alveolar cytoplasm of type I cells can be seen scattered in small vacuoles;Besides,the microvilli of the free surface of the alveolar cells of the type II were partly lost and existed in disorder,at the same time,some lamellar bodies were vacuolated,and mitochondria swelled,as well as uneven distribution of chromatin.And the thin layer of connective tissue and basement membrane were thickened.In a word,compared with the model group,all of the above situations were improved.Conclusion:1.Quanzhenyiqi decoction has the effect of increasing the weight of COPD mice,protecting pulmonary function,improving the lung tissue pathology and ultrastructure of mice.2.Quanzhenyiqi decoction can reduce the number of inflammatory cells in bronchoalveolar lavage fliud,reduce the expression of IL-6 and IL-8,and relieve the airway inflammation of COPD mice.3.It can be supposed that Quanzhenyiqi decoction play an anti-inflammatory effect through decreasing the expression of ERK and Nrf2 protein,while increasing the expression of HO-1 protein.
