Objective:To investigate the expression of serum brain-derived neurotrophic factor(BDNF)in children with asthma,and to explore the possible role of BDNF in the pathogenesis of asthma in children and its relationship with the severity of asthma.Methods: From July 2015 to July 2016,60 cases of children with acute asthma(asthma group)and 60 healthy children(control group)were chosen as the research object in the department of Pediatrics of the First Affiliated Hospital of Shihezi University Medical College,and the basic clinical data of children with asthma were collected.They were divided into mild group with 18 cases,moderate group with 25 cases and severe group of 17 case according to the severity of asthma,including 39 cases of male,female 21 cases,age 1.3~14.0 years old,the average was(8.1±3.2)years;in the control group,38 cases were male,22 were female,aged1.1~14.4 years old,the average was(7.4±3.2).collected fasting venous blood of 2ml before treatment and symptom remission(no wheezing symptoms,without pulmonary wheeze,standardized treatment control)of children were chosen;and collected fasting venous blood2 ml of the control group;with room temperature(20~25 ?)static 60 min,3000r/min centrifugal the serum with 10 min,retained for inspection in the condition of-70 ? after packed in EP tubes,refrigerator to avoid repeated freezing and thawing.The application of enzyme linked immunosorbent assay(enzyme-linked immunosorbent,assay,ELISA)two There is no difference between the expression level of BDNF in serum of children group,difference between the severity of asthma children serum BDNF level,and analyze the relationship between BDNF level and the severity of asthma in children,and basic clinical data were statistically analyzed.The data was statistically analyzed by SPSS 17.0 statistical software.Measurement data to the mean and standard deviation(x ± s),obey the normal distribution and variance using t test or analysis of variance,homogeneity of variance using non parametric test;count data to rate expressed by chi square test;the correlation between the two variables by Correlation analysis.With P<0.05 as the difference was statistically significant.Results:1.Serum levels of BDNF group of acute asthma symptoms and group were(8.8±0.6)ng/ml,(6.4±0.9)ng/ml,and were significantly higher than the control group(1.9±1.2)ng/ml,with acute exacerbation group was the highest(P<0.05),BDNF level of symptom remission was lower than the acute stage,the difference was statistically significant(P < 0.05).2.The serum level of BDNF were different with asthma severity in children in groups: mild minimum(8.2±0.4)ng/ml,severe group was the highest(9.5±0.3)ng/ml,the difference was statistically significant(P<0.05).The higher the severity of asthma,the higher the expression level of serum BDNF,the positive correlation between the two(r=0.756,P<0.05).3.33 cases of asthma group had atopic constitution,and the other 27 cases had no specific constitution,the serum BDNF levels of both groups were(9.1 ± 0.6)ng/ml,(8.4 ± 0.5)ng/ml.The results showed that the level of serum BDNF in asthmatic children with atopic constitution was higher,the difference was statistically significant(P<0.05).Correlation analysis showed that there was a positive correlation between them(r=0.504,P < 0.05).Conclusion: BDNF may play an important role in the pathogenesis of asthma in children,it may be related to the atopic constitution of children,and the expression level of serum is related to the severity of asthma.The detection of serum BDNF level will be helpful to evaluate the condition of children with asthma,and may have a certain effect on the development of asthma.
