| Objective:To investigate the mRNA and protein expression of STIM1 in Kaposi`s sarcoma(KS),and explore the relationship of them with KS’S clinical and pathological,then,primary study results on the role of STIM1 in the pathogenesis of KS,provide the necessary theoretical and experimental basis for further study of new treatment target.Methods: Real-time fluorescent quantitative PCR and immunohistochemistry were performed to measure the mRNA and protein expression of STIM1 of paraffin-embedded KS,hemangioma and normal skin tissue specimens,observe and complate their difference.Analysis the expression of STIM1 in KS with different age,sex,national,pathological types,as patch phase,plaque phase and nodular phase,and HHV-8 infection,HIV infection.Using the SPSS17.0 software to detected the expression data of STIM1 for statistical analysis.Results: 34 patients with a diagnosis of Kaposi’s sarcoma,including 24 classic and 10 AIDS-Kaposi’s sarcoma.Male-to-female ratio: 29∶5.Mean age at diagnosis was(56.55±16.07)years.The main types of lesions were nodules(18 cases),patches(5 cases),and plaques(11 cases).The area of lesions was 22 cases(≤ 5%),12 cases(≥ 5%).The relative expression of STIM1 mRNA in the tissue of KS,hemangioma,normal skin tissue are(1.290 ± 1.215),(1.546 ± 1.106)and(1.321 ± 0.964),there are no significant differences in the three groups(p < 0.05),as in KS’s different sex,national,pathological types,HHV-8 infection,HIV infection and pathological staging(p>0.05).While,the protein of STIM1 was detected in 27(79.41%)of 34 paraffin-embedded KS,the positive expression rate in hemangioma and normal skin tissues were 81.81% and 13.33%,with a significant difference between KS and normal skin(p <0.05),but not in KS and hemangioma.Also,the protein expression of STIM1 in paraffin-embedded KS tissues was no correlation with ethnic,sex,HHV-8 infection,HIV infection,skin lesions area(p > 0.05),as between the patients positive with both HIV and HHV-8 and negative with both HIV and HHV-8(Z=-0.444,p>0.05).But,the positive expression of STIM1 is positively correlated with KS`s pathological classification(r=0.611,Z=0.000),nodular phase is higher than plaque phase(Z=0.068,p<0.05)and patch phase(Z=-2.323,p=0.02).Further analysis the correlation of STIM1 mRNA and protein expression level showed negative correlation(r =-0.058).Conclusion :STIM1 in kaposi’s sarcoma tissues all are expressed with different degrees,the expression of mRNA level showed no difference compared with normal tissues,while the protein expression was significantly high,which preliminary clarify that he post-transcriptional regulation of STIM1 may affect the process of the pathological histology of KS.There was no difference between kaposi’s sarcoma tissues and hemangioma in STIM1 expression,and with the pathological staging of kaposi’s sarcoma tissues were positively correlated,nodular stage is highest,as vessel is obvious growth in nodules blood,which suggests that STIM1 is probably promote the process of the pathological histology of kaposi’s sarcoma by promoting blood vessel growth. |
