Objective:The genotype distribution and gene frequency distribution of A79 C and G208 A loci in Xinjiang Uygur and Han NSCLC were detected by detecting the polymorphisms of CDA A79 C and G208 A loci in peripheral blood of Xinjiang Uygur and Han NSCLC.And to explore the relationship between gene polymorphism and chemotherapeutic efficacy in NSCLC patients,and to provide theoretical basis for individual therapy of gemcitabine in patients with NSCLC.Methods: Collected from August 2014 to April 2016 in Shihezi University Medical College First Affiliated Hospital of Cancer and Kashi District First People's Hospital by the Pathology of advanced NSCLC newly diagnosed Uygur patients in 53 cases,67 cases of Han patients The gene polymorphisms of A79 C and G208 A were detected by direct sequencing.All patients with NSCLC were treated with Gemcitabine-containing chemotherapy.After two cycles of chemotherapy,the differences of gene polymorphism distribution between the two groups were analyzed.Results:(1)The frequencies of CDA A79 C and G208 A genotypes in 53 Uygur and 67 Han patients with NSCLC were in accordance with Hardy-Weinberg equilibrium(P> 0.05).(2)There were no significant differences in the clinical data between the Uygur and Han NSCLC patients in Xinjiang,and the frequency of the mutant allele was 47.8%,the mutation allele frequency was 32.8%,the Uygur(P <0.05).The frequency of mutant allele was 24.6% and the mutation allele frequency was 5.1%.The frequency of allele and allele were statistically significant(P <0.05).The frequency of mutant genotype was 22.4%,the mutation allele frequency was 11.2%,the frequency of mutant genotype in Uygur was 5.7%,and the mutation allele was found in CDA G208 A.Frequency was 2.8%,and the genotype frequency and allele frequency were statistically significant(P <0.05).(3)In the AC genotype of the A79 C gene,the effective rate of the Han was 35% and the Uygur effective rate was 30%.The clinical efficacy of the Han and UG ethnic groups was statistically different(P = 0.028).In the genotype of G208 A GG genotype,the effective rate of Han was 55.8%,the Uygur effective rate was 32%,and the clinical curative effect of Han and UG ethnic group NSCLC was statistically different(P = 0.016).(4)The positive rate of AA,AC and CC genotype of A79 C gene in Han patients with NSCLC was 65.7%,35% and 25% respectively.There was significant difference between the two groups(P = 0.016).The positive rates of AA,AC and CC genotype of A79 C gene in Uygur ethnic group were 62.5%,30% and 0 respectively,and the difference was statistically significant(P = 0.031).Conclusion: 1.In NSCLC patients,there were racial differences in the genetic polymorphisms of the two sites of CDA A79 C and G208 A in Uygur and Han nationality.2.The efficacy of A79 C gene AC genotype and G208 A gene GG genotype in patients with non-small cell lung cancer were superior to those of UG ethnic group.3.The polymorphism of A79 C in patients with NSCLC is closely related to the effect of chemotherapy,and the efficacy of AA genotype chemotherapy is superior to that of AC and CC genotypes,which may be related to the chemotherapy of patients with NSCLC in the future Potential application value,for the clinical individualized drug reference.
