Background:Polycystic ovary syndrome(PCOS)is one of the most frequent endocrine and metabolic disorders,and it has been associated with obesity.The pathogenesis of PCOS is still unclear,and it is considered to be caused by genetics,environment and other risk factors.Micro RNAs(miRNAs)are a class of small endogenous non-coding single-stranded RNA molecules that regulate gene expression at the post-transcriptional level.Abnormal expression of miRNAs has been related to several metabolic disorders including obesity,diabetes,and PCOS.Recently many scholars found that miR-21,miR-27 a,miR-27 b have been associated with inflammatory processes,hormone metabolism,adipogenesis,obesity,and insulin signaling.However,there are little research on the relationship between the level of miR-21,miR-27 a,miR-27 b and obesity,metabolic disorders in PCOS.Objective:To investigate the expressions of miR-21,miR-27 a,miR-27 b in peripheral blood lymphocytes of PCOS with obesity group,non-obesity group and the effect of weight control on them.Then explore the correlation between miR-21,miR-27 a,miR-27 b and obesity,metabolic disturbance(hyperandrogenism and hyperinsulinemia)in PCOS.Methods:1.60 patients with PCOS were divided into 2 groups according BMI.22 cases with BMI<25kg/m2 were non-obesity group,38 cases with BMI?25kg/m2 were obesity group.2.The levels of b FSH,b LH,b E2,b P,b T,FINS were determined by using the electrochemiluminescence method.3.The levels of Glu were detected by the glucose oxidase method.4.The levels of miR-21,miR-27 a,miR-27 b in lymphocytes were detected by quantitative real-time polymerase chain reaction.5.The above indexes were checked again after controlling weight in obesity group.Result:1.Compared with the non-obesity group,the levels of b T,Glu,FINS,HOMA-IR in obesity group were significantly higher(P<0.05).2.The levels of miR-21,miR-27 a,miR-27 b in obesity group were higher than those in the non-obesity group(2.83±0.60 vs 1.18±0.24;2.63±0.42 vs 1.13±0.19;2.74±0.49 vs1.16±0.17;P<0.05).3.The levels of b FSH,b E2,b P had no obvious difference(P>0.05),while the levels of BMI,b LH,b T,Glu,FINS,HOMA-IR were significantly decreased(P <0.05)after controlling weight in obesity group.4.The levels of miR-21,miR-27 a,miR-27 b in obesity group were significantly decreased after controlling weight(1.88±0.27 vs 2.81±0.61;2.16±0.39 vs2.69±0.37;2.27±0.40 vs 2.72±0.47;P<0.05).5.The expression of miR-21 was positively correlated with b T and the expressions of miR-27 a,miR-27 b were positively correlated with FINS.Conclusions:Mi R-21,miR-27 a,miR-27 b are closely related to obesity and metabolic disturbance(hyperandrogenism and hyperinsulinemia)in PCOS.miR-21 may be mainly associated with the abnormal metabolism of b T,and miR-27 a,miR-27 b may be mainly associated with the abnormal metabolism of insulin.miR-21,miR-27 a,miR-27 b are involved in the vicious circle of obesity,yperandrogenism and hyperinsulinemia in PCOS through different routes.
