Objective:To investigate the m RNA and protein expression levels of ITGA3 with real time PCR and immunohistochemistry stainning in epithelial ovarian cancer(EOC)and analyze the correlation between the expression of ITGA3 and clinical characteristics of patients with EOC.Methods:(1)Using real-time quantitative PCR to detect the m RNA expression level of ITGA3 in different EOC cell lines,normal epithelial cell line,EOC tissues and benign epithelial ovarian tissues.(2)The expression of ITGA3 protein in EOC tissues and benign ovarian tissues was detected by immunohistochemistry(IHC).The 27 ovarian tissue specimens from patients with benign ovarian disease must be ovariectomized and 38 ovarian tissue specimens from patients with EOC were obtained from the affiliated hospital of Jiangsu university,the affiliated people's hospital of Jiangsu university and the second people's hospital of Lian Yungang between the year of 2013 and 2016.The clinic data of these patients were also collected and analyzing the relationship between the expression of ITGA3 protein and clinical characteristics of epithelial ovarian cancer.Results:(1)The m RNA expression levels of ITGA3 in SKOV3 and OVCAR-3 two ovarian cancer cell lines were higher than in normal epithelial ovarian cell line Hosepic(P<0.001).(2)The m RNA expression levels of ITGA3 in EOC tissues were significantly higher than in benign ovarian tissues(P<0.001).(3)The results of IHC show that ITGA3 protein were both expressed in benign ovarian tissues and EOC tissues.There were 26/38 positive specimens in EOC tissues but only 4/27 positive specimens in benign ovarian tissues(P=0.000).The higher expression of ITGA3 in advanced stage was more obvious in EOC tissues(P=0.016,P=0.003).Conclusion: ITGA3 may be regarded as a marker for judgment EOC malignancy and provide an evaluation for clinical therapy and may be a potential therapeutic target.
