Objective Interleukin 8(IL-8)and interferon-gamma(IFN-?)are important factors associated with cellular immunity.To detect IL-8 and IFN-? in cerebrospinal fluid of tuberculous meningitis(TBM)in this study,and to explore the significance of dynamic chages of IL-8 and IFN-? in cerebrospinal fluid in diagnose,possible mechanismsin and prognosis of TBM.Methods Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of IL-8and IFN-? in the CSF of patients with tuberculous meningitis(n=30),viral meningitis(n=30)and noninfectious neurologic diseases(n=30)who admitted to the second hospital of lanzhou university from December 2014 to August 2016 according to the corresponding clinical diagnostic criteria.According to the TBM staging criteria proposed by Medical Research Council,12 patients of stage I,11 patients of stage II,7 patients of stage III.To detect the levels of IL-8 and INF-? in CSF from TBM patients,viral meningitis patients,noninfectious neurologic diseases patients and the TBM patients after 1 weeks' and 2 weeks' treatment in this study.Statistics analysis was performed with SPSS18.0.The comparison between the three groups using Single factor repetitive measure analysis of variance and comparing for paired samples T test within the group,and the dependency between cytokines level and the stage of TBM by using Pearson correlating analysis.Results(1)CSF levels of IL-8 in TBM group were(4335.5±1235.4)pg/ml,which were significant higher than the(121.483±50.062)pg/ml in VM group(P<0.05)and(22.980±16.940)pg/ml in noninfectious neurologic diseases group(P<0.05).CSF levels of IFN-? in TBM group were(52.9±12.5)pg/ml,which were significant higher than the(25.467±7.039)pg/ml in VM group(P<0.05)and(8.173±2.609)pg/ml in noninfectious neurologic diseases system(P<0.05).Moreover,CSF levels of IL-8 and IFN-? in control VM group were significant higher than noninfectious neurologic diseases group(P<0.05).(2)CSF levels of IL-8 and IFN-? of TBM group were(4335.5±1235.4)?(52.9±12.5)pg/ml before treatment,which were significant higher than(2798.4±1109.6)?(40.2±9.7)pg/ml of 1 weeks' treatment(P<0.05)and(2246.7±1010.6)?(33.7±9.5)pg/ml of 2 weeks' treatment(P<0.05).Moreover,the levers of IL-8 and IFN-? in TBM group before treatment might be correlated with the disease stages.Conclusion The levels of IL-8 and IFN-? in anti tuberculosis treatment continued to decline,which were positively related to the TBM stages.The more higher levels of IL-8 and IFN-?,the more serious condition of the patients,which further indicated that the levers of IL-8 and IFN-? of great significance for the pathogenesis of TBM.Joint detection of IL-8 and IFN-?in cerebrospinal fluid has certain clinical application value in diagnosis tuberculous meningitis,and the levels is related to the severity of the disease.The dynamic change of IFN-? and IL-8 in CSF can predict the prognosis of TBM.Therefore,the levels of IFN-? and IL-8 of CSF has certain clinical significance for early diagnosis of TBM,which can also dynamic observation the treatment effect of TBM.As potential diagnostic indicators of TBM,IFN-? and IL-8 are contribute to reducing the mortality and disability and improve the prognosis.
