Objective:To investigate the pharmacokinetics characteristics of propofol in pigs with volume controlled hemorrhagic shock in high altitude.Methods:Twenty-four Landrace–Large White pigs were randomly divided into 4 groups: group C(swine without hemorrhagic shock in high altitude),group HS(swine with hemorrhagic shock in high altitude),group JR(swine with hemorrhagic shock and crystalloid solution resuscitation),group HR(swine with hemorrhagic shock and colloid solution resuscitation).The hemorrhagic shock models were established.Propofol 150?g·kg-1·min-1was infused for 10 min;2 ml blood was collected before into of propofol and at 1,2,3,4,10 min,after infusion at 1,2,4,8,15,25,35,50,70,90,120,150,180 min,and the concentration of propofol was analyzed by high performance liquid chromatography ultraviolet(HPLC).SPSS was used to calculate the elimination half-life(t1/2)of propofol,the drug's area under the curve(AUC),the plasma-effect site equilibration rate content(Ke0),the mean residence time(MRT)and the other pharmacokinetic parameters.Results:The plasma concentration of propofol in group HS and group JR were higher than that of group C,there being significant differences(P<0.01),in group HR was not higher than those of group C,there being no significant differences(P>0.05).Cmax in group HS and group JR were higher than that of group C,there being significant differences(P<0.01),in group HR was not higher than those of group C,there being no significant differences(P>0.05).The t1/2 and MRT in group HS and JR were longer than group C,there was significant differences(P<0.05),in group HR was not higher than those of group C,there being no significant differences(P >0.05)The Ke0 and AUC in group HS and group JR were higher than those of group C,there was significant difference(P<0.05),in group HR was slightly higher than those of group C,but there was no significant difference(P>0.05).Conclusion:The pharmacokinetics characteristic of propofol in swine with volume controlled hemorrhagic shock in high altitude is manifested as slower metabolism,longer elimination half-life,increased Ke0 and longer MRT.The same amount of crystalloid solution resuscitation was not enough to improve the low blood volume,cause of propofol metabolism slows,blood drug concentration increased,and the same amount of colloid can reverse the pharmacokinetics of change.
