Objective To observe the effect of Xiao Banxia Decoction on gastric emptying and small intestinal propulsion in mice and on the movement of isolated ileum in rat,and to explore its effect on gastrointestinal motility and its mechanism.Methods 1.To establish the model of gastrointestinal motility in mice by cisplatin,neostigmine,atropine,adrenaline,and dopamine.The effect of Xiao Banxia Decoction on the gastrointestinal motility of normal mice and model mice was observed by measuring the residual rate of stomach and the rate of small intestine propulsion.2.Using acetylcholine,barium chloride,dopamine,adrenaline and atropine as tools,we observed the effects of different concentrations of Xiao Banxia Decoction on the contraction of isolated ileum in rats.Results 1.Experimental results of gastric emptying and small intestinal propulsion:It was shown that XBXD significantly antagonized neostigmine induced gastrointestinal motility hyperfunction.XBXD can antagonize atropine-induced gastric emptying inhibition,not the small intestinal propulsion inhibition.It also significantly antagonized dopamine and adrenaline induced gastric emptying inhibition and small intestinal propulsion inhibition.2.Experimental results of isolated ileum:XBXD could dose-dependently inhibit ileum spontaneous contractions,reducing the average tension,amplitude.All the three doses(20,10,4g·L-1)of XBXD could suppress the contractility,significantly reduced the tone and amplitude of ileum contractions stimulated by Acetylcholine(ACh)and Barium chloride(Ba Cl2).All the three doses had additive effects on tone and amplitude of ileum contractions with dopamine(DA).All the three doses had no effects on the inhibition of ileum contraction induced by atropine and adrenaline.Conclusion 1.XBXD can antagonize neostigmine-caused gastric emptying and small intestine hyperfunction,while improving atropine-induced gastric emptying inhibition,antagonizing dopamine and adrenaline-induced gastric emptying and small intestine propulsion.Suggesting that XBXD has a two-way regulation of gastrointestinal motility.2.XBXD have an inhibitory effect on rat ileum contractions and increase with the increase of dose.It can antagonize the contraction of ACh and Ba Cl2 induced intestinal contraction in rats,and play the role of DA in the relaxation of intestinal tract.This study shows that XBXD can regulate gastrointestinal motility.The mechanism might be mediated by cholinergic receptors,adrenergic receptors,dopamine receptor systems,or by direct action on smooth muscle.The mechanism of action of XBXD on gastrointestinal motility needs further study.
