Objective1. To observe the effect of Xiaopi No.1 using mouse gastric emptying and small intestinal pushing experiments.2. To observe the effect of Xiaopi No.1 to rabbit isolated gastric muscle strip and intestinal duct, and the further mechanism.Method1.The healthy mice have been divided into different groups according to the weight at random. There were 15 mice in each group and they were treated by gastric irrigation for one week. After the mice were fasting for 24 hours, they were irrigated by nutritional semisolid paste. In this condition, after 20 minutes, the effects of Xiaopi No.1 to the functions of gastric emptying and small intestinal pushing in normal mice with the rates of gastric emptying and small intestinal pushing.2.The models of mice with decreased gastric emptying and small intestinal pushing rates by using atropin, dopamine and ephedrine were made and the rates of gastric emptying and small intestinal pushing were also detected as method 1. The rates of gastric emptying and small intestinal pushing were observed after 0 minutes of the last time injection of atropin, dopamine or ephedrine to the abdomen.3. The mice were divided by weights and grouped at random. There were 10 mice in each group and they were treated by gastric irrigation for one week. Then the mice were fasting for 24 hours. After the mice were injected by different drugs as above, 4ml femoral vein blood were sampled and the GAS and MTL were detected.4.10 healthy rabbits were fasting for 24 hours and immediately executed. Then the gastric strips, sinus circle piece, pylorus circle piece, middle parts of small intestinal muscle and colonic muscle were got rapidly. All the samples were kept in Krebs tank with constant temperature 37℃, added Xiaopi No.1 in different concentrations and observed the average shrink extent, frequency and strain.Result1. The use of Xiaopi No.1 in low, middle or high concentration and also CIS could obviously increase the gastric emptying rate of normal mice. There were significant difference compare with the control with distilled water. Meanwhile, the use of middle or high doses of drugs had the effects to the promotion of small intestinal pushing in normal mice, but there were no visible dose-effect relationship for the gastro-intestinal motor.2. Xiaopi No.1 could resist the effect of gastric emptying and small intestinal pushing delay caused by atropin and dopamine (P<0.05, P<0.01), but couldn't resist those by ephedrine.3. Xiaopi No.1 could enhance the tension and average swing of rabbit gastric strips (P<0.05, P<0.01), but no difference to the constriction frequency (P>0.05). It also could decrease the motor index of sinus circle piece, the tension, average swing and constriction frequency of intestinal tube. There were so difference found in the effects of tension, swing and constriction frequency to sinus strip and intestinal tube.Conclusion1. Xiaopi No.1 can significantly increase the function of gastric emptying and small intestinal pushing. It can accelerate the gastro-intestinal motor. There should have relation of this effect by interdiction of M receptor and dopamine receptor but not the adrenergic receptor and also the gastric sympathetic. Xiaopi No. 1 also can increase the serum level of MLT and GAS.2. Xiaopi No. 1 can significantly increase the gastric smooth muscle tension and average swing of rabbit. It also can decrease the motor index of sinus circle piece, the tension, average swing and constriction frequency of intestinal tube. There were clear changes in the effects of tension, swing and constriction frequency to sinus strip and intestinal tube. So it's possible that the enhancing effects come from the direct stimulation of drug to gastro-intestinal smooth muscle and also the increased secrete of gastro-intestinal hormone. |
