Objective: Study on the expressions of mRNA in NEU1 and LAMP-1 in hepatic tissue of mouse based on immune thrombocytopenia model by using the single herb such as the herb of Hairyvein Agrimonia;discuss the relations of ITP with the expression degree of NEU1 and LAMP-1,and analyze the correlations of pathogenesis of NEU1 and LAMP-1 with ITP,thus to seek new ideas for the research and curing.Methods: 1.Utilize the intraperitoneal injection modeling method by injecting APS serum to BALB/C mouse,and establish the immune thrombocytopia model in mouse;detect the blood platelet number in peripheral blood,the total number of bone marrow megakaryocytes and the number of megakaryocyte production board,thus to comprehensively evaluate the model.2.Utilize the random approach to separate 60 BALB/C mice into 5 groups,including nomarl control group,model control group,herb of Hairyvein Agrimonia group,adenophora trachelioides maxim group,and herba sarcandrae group,and compare the normal conditions,the pathological morphology of related tissues,the number of blood platelet in peripheral blood and the morphology of bone marrow cells of mice in each group before and after herb intervention.3.Apply RT-QPCR and Western-blotting method to detect the expressions of mRNA in NEU1 and LAMP-1 in hepatocyte in each group of mice,and probe into the relations of ITP with NEU1 and LAMP-1,as well as the effects of herb intervention on immune thrombocytopenia.Results: 1.After applied intraperitoneal injection APS method to build animal model,the blood platelet apparently decreased in peripheral blood,and bone marrow megakaryocyte was inhibited to be mature,which was accorded with ITP clinical features of human being,thus this model can be applied to study on ITP animal immune model.2.The intervention of herb of Hairyvein Agrimonia,adenophora trachelioides and herba sarcandrae can apparently improve the general conditions of ITP model mice,and increase the number of blood platelet and bone marrow megakaryocyte with great difference(P<0.05).3.As RT-QPCR results showed,the expression degree of mRNA greatly increased in NEU1 and LAMP-1 gene in modeling group compared to normal group(P<0.01);the increasing degree of mRNA expression in NEU1 and LAMP-1 genes decreased at different degrees in herb of Hairyvein Agrimonia group,adenophora trachelioides Maxim group and herba Sarcandrae group(P<0.05).4.The detection results of Western-blotting and RT-QPCR were basically similar,and in model group,the expression of NEU1 at protein degree increased apparently(P<0.01).Conclusion: Both the NEU1 and LAM-1 coded protein can closely correlate with the normal participation of lysosome and the cell function of exocytosis,devour,autophagy and immune;the up-regulated expression of these two genes may be closely related to IP pathogenesis,and play important roles in the destruction of blood platelet;the three types of herb such as herb of Hairyvein Agrimonia,adenophora trachelioides and herba sarcandrae which are often used on clinic can inhibit the desialylation of NEU1 at different degree,but the specific effect and mechanism need to be further researched.
