Prescription Of Reduce Stasis And Protect Liver On The Treatment Of Severe NAFLD In Rats And Mechanism

Objective:1.To establish the model of moderate and severe NAFLD,we use high fat diet and low dose of carbon tetrachloride(CCL4)soybean oil.By this means,we can get a rapid prototyping,low risk and easy replication model.2 Compare with low dose group,high dose group of Prescription of reduce stasis and protect liver and Silibin Meglumine Tablets' s efficacy and pharmacology,choose different treatment methods.Methods: 1.Using 60% high fat diet combined with low dose of carbon tetrachloride(CCL4)method of soybean oil solution,6,8,12 to observe the general conditions of rats,liver function,blood lipid and pathology,were selected for modeling time.2.Use low / high dose of prescription of reduce stasis and protect liver and common dose of Silibin Meglumine Tablets in the treatment of moderate to severe NAFLD rats.TG,TC,HLD-C,LDL-C,ALT,AST,FFA,TNF-? and TNF-?1,MAD,SOD,CYP2E1 can be detected.We observe the degree of hepatic steatosis,inflammatory cells infiltration,mining treatment efficacy,mechanism analysis,and to determine the optimal dose.Results: 1.Simple hepatic steatosis was established with high fat diet combined and low dose of CCL4 on the weekend of 6 to establish,which was completed by the end of 8 weekends.The 12 weekend prompted establish moderate and severe NAFLD.2.Through 4 weeks of treatment,(1).In the aspect of improving liver function,each group of treatment can improve liver function ALT,AST effect(P<0.05),high dose group has better effect,three groups have no significant reduction of FFA(P>0.05).(2).In improving blood lipids,each group of treatment TC,LLD-C decreased obviously(P<0.05),silymarin group better effect in reducing TG,low and high dose groups had no significance(P>0.05),silymarin decreased obviously(P<0.05),(3).In antioxidant aspect,three groups decreased obviously effect on MDA(P<0.05),silymarin has better effect.Low dose group increased significantly(P>0.05),high dose group and silymarin group increased(P<0.05),SOD is better in reducing inflammatory appearance,(4).In terms of reducing inflammatory factors,low dose group and silibinin group decreased levels of TNF-?(P>0.05)is not obvious,the effect of high dose group significantly(P<0.05).The low dose group of TNF-?1 level is not obvious,the effect of high dose(P>0.05)and silymarin group significantly(P<0.05).In terms of reducing CYP2E1,low dose group of TNF-beta 1 level is not obvious,the effect of high dose(P>0.05)and silymarin group significantly(P<0.05).Conclusion: In our study,we established a simple and easy to replicate NAFLD rat model.In treat severe NAFLD,Prescription of reduce stasis and protect liver has significant effect,the mechanism of the prescription can reduce inflammation,protect the liver function,reduce fat content.The high dose of rescription of reduce stasis and protect liver can reduce inflammatory,protection of liver function and reduce fat content.Among them,the high dose of the prescription has better effect in antiinflammatory and protect liver.Silibin Meglumine Tablets has better effect in lipid-lowering and antioxidant.
