Theeffect Of Zhi Gan Prescription On Theregulation Of NF-?B-TNF-?/Bcl-2 Pathwayin Nonalcoholic Fatty Liver Disease

Objective: The subject focuseson the function of signal channel“NF-?B-TNF-?”/ Bcl-2on nonalcoholic fatty liver disease by building a rats model carrying nonalcoholic fatty liver disease induced by high-fat diet.Also,it aims to explore “NF-?B-TNF-?” / Bcl-2 interventional effect of hepatocyte inflammation and apoptosis Zhi Gan prescription in nonalcoholic fatty liver disease mediated signaling pathways.Methods: Eighty SD rats are randomly allocated to Blank group and Normal control group and some other group.Ten rats in the former group will be put to death in 8 weeks,and ten in the normal group is fed with standard diet before put to death in 12 weeks.After 8 weeks' high-fat diet,10 rats are picked out from the 60 rats in random as 8-weeks Model group,which will be put to death in 8 weeks.The other 50 will be kept fed in high-fat diet until the end of the experiment,and randomly divided into 12-weeks Model group,low-dose Zhi Gan Prescription group,mid-dose Zhi Gan Prescription group,high-dose Zhi Gan Prescription group and Western medicine group,10 in each.Make intervention after the 8th weeks till the 12 th weeks,during which give a gavage of Zhi Gan prescription in low,mid,high dose to the rats in the Chinese Medicine group(Zhi Gan Prescription group),and a gavage of Atorvastatin Calcium Tablets to the Atorvastatin Calcium Tablets.After 12 weeks,rats in each group are fasting before put to death to get blood.Separate the serum by the help of automatically biochemical instrument and then to realize the following object: the serum level of ALT?AST?TG?TC;Observing the pathological alteration of the hepatic tissue in each group in HE staining;Western Blot detection protein expression NF-?B?bcl-2 and TNF-?level.Results: The experiment shows that the serum ALT?AST?TG?TC in Model group are obviously higher than the Blank group of the same period(P<0.01).Also,the degrees of inflammatory cells' infiltration,necrosis and liver steatosis in 12-weeks Model group are obviously more serious than the ones in 8-weeks Model group(P<0.05).Compared to the Model group,Treatment group'sfatty inflammatory and lipid accumulation are obviously decreased and improved according to the pathology,for which the rats' liver tissue pathology changes and tends to be normal.What's more,the content of serum TG,TC,and the ALT and AST of liver functionare inducted in the treatment group.On the function of decreasing serum TC,TGand ALT,AST,high-does Zhi Gan prescription is the greatest.The result of Western Blot shows that the NF-?B protein expression level is higher in the model group than the blank group at same period.However,bcl-2 protein expression level in model group is apparently lower(P<0.05).Compared to the model group of same period,NF-?B protein expression decreases apparently in the treatment group(P<0.01),while its bcl-2 protein expression level increases(P<0.01).And the high-does Zhi Gan prescription is the best in the treatment group.The same period compared with the blank group,model group liver tissue TNF-? expression significantly increased(P<0.01),after drug intervention groups ofTNF-? reduced expression to different extent,high dose group is superior to other treatment group(P<0.01).Conclusion(s): Zhi Gan prescription can protect liver,for it can not only improve liver steatosis of the experiment rats with nonalcoholic fatty liver but also regulate lipid metabolism and restrain lipid droplet to assemble.Zhi Gan prescription lighten inflammatory response by decrease the level of NF-?B andTNF-?protein expression,and restrain liver apoptosis by bcl-2 up-regulation.The experiment indicates thatZhi Gan prescription has the function of anti-apoptosis and can lighten inflammatory response of nonalcoholic fatty liver disease as well.
