Expression Changes And Clinical Significance Of Ezrin And P70S6K In Three Bone Tumors

By testing the expression of Ezrin and P70S6 K in the Osteosarcoma(OS),giant cell tumor of bone(GCTB)and(osteochondroma,OC)tissues,to study the expression differences among them respectively and analyse the relationship between the expressions of Ezrin and P70S6 K and the Clinical pathological features of OS and GCTB,and the correlation between Ezrin and P70S6 K in OS and GCTB,so can providea theory evidence for the research on invasion,metastasis and recurrence mechanism of OS or(and)GCTB,and also a new target for the diagnosis and treatment of OS and GCTB.All 100 samples of 40 cases with OS,40 cases with GCTB and 20 cases with OC were collected from the Department of Pathology of the 251 st Hospital of PLA from January 2000 to January 2015.All patients before sampling without any treatment and exclude that who suffers from any other underlying disease.The diagnosis of bone neoplasms should be based on a combination of clinical evaluation,pathological laboratory and radiographic options.The expression of Ezrin and P70S6 K protein were detected in three tissues by immunohistochemistry(IHC).To study the expression differences among them respectively,and the relations between the two markers and the gender,age,tumor position,tumor size and pathological type,Enneking staging,differentiation degree,softtissue invasion,pulmonary metastasis of OS and also the gender,age,tumor position,Jaffe grading,three combined stage,soft tissue invasion,recurrence or metastasis of GCTB were analysed,respectively.The correlation between the Ezrinand P70S6 K in OS and GCTB was analysed by Spearman rank correlation as well.Positive staining results of Ezrin and P70S6 K are all a light yellow,tan or brown calm in the tumor cells.Ezrin is mainly located in the cytoplasm.The positive expression rates of Ezrin in OS,GCTB and OC were 72.5%(29/40),42.5%(17/40),15.0%(3/20)respectively.P70S6 K is mainly located in the cytoplasm.The positive expression rates of P70S6 K in OS,GCTB and OC were 67.5%(27/40),40.0%(16/40),10.0%(2/20)respectively.The expression of Ezrin and P70S6 K in OS were not correlated with the gender,age,tumor position,tumor size and pathological type(P>0.05;P>0.05),but correlated with Enneking staging,differentiation degree,soft tissue invasion and metastasis(P<0.05;P<0.05).The expression of Ezrin had a positive correlation with P70S6 K in OS(r=0.409,P<0.05).The expression of Ezrin and P70S6 K in GCTB were not correlated with the gender,age,tumor position(P>0.05;P>0.05),but correlated with the Jaffe grading,three combined stage,soft tissue invasion,recurrence or metastasis(P<0.05;P<0.05).The expression of Ezrin had a positive correlation with P70S6 K in GCTB(r=0.434,P<0.05).There were significant differences of positive expression rates of Ezrin and P70S6 K between OS and OC(P<0.001,P<0.001).There were significant differences of positive expression rates of Ezrin and P70S6 K between GCTB and OC(P<0.05,P<0.05).There were significant differences of positive expression rates of Ezrin and P70S6 K between OS and GCTB(P<0.05,P<0.05).Both Ezrin and P70S6 K are highly expressed in OS and GCTB,suggesting that they may play important roles in the occurrence,development,invasion and metastasis or recurrence of OS and GCTB.The positive correlation between the expression of Ezrin and P70S6 K in OS and GCTB,suggesting that Ezrin and P70S6 K may have a synergistic interaction and participate in the pathogenesis of OS andGCTB together.So,Ezrin and P70S6 K can be a new target for the study of diagnosis and treatment of OS and GCTB.The positive expression rates of Ezrin and P70S6 K in OC,GCTB and OS are increase in turn,suggesting that Ezrin and P70S6 K may predict the benign and malignant of bone tumors effectively.
