Objective:Sprague Dawley rats in vivo model were used to study the cardioprotection of dexmedetomidine.To compared with the infarction regions between samples to explore the cardioprotection of dexmedetomidine.Microarray was used to analyze miRNA differentially transcriptome.Methods:Healthy Specific Pathogen Free male Sprague Dawley rats,weighing 250-300g were used in our study.Sprague Dawley rats were divided into four groups randomly:control group(group C),ischemia/reperfusion group(group I/R),ischemia preconditioning group(group IP)and dexmedetomidine preconditioning group(group DP).Group C was perfused with blood continuously for 180min:The hearts in group IR were ischemia 30min and then continued perfusing 120min;In the group DP,dexmedetomidine(20ug/kg)was given via the right internal jugular venous 15min before ischemia;Group IP was ligation of the left anterior descending artery 5min and then reperfusion for 5min,three cycles(ischemia preconditioning).Group IP and group DP were subjected to 30 minutes of global ischemia followed by 120 minutes of perfusion.TTC dying method was used for infarct size determination.Microarray was used to analyze miRNA differentially transcriptome.Results:Compared with other groups,the myocardial infarction size of group C was significantly decreased(P<0.05).Compared with group IR,myocardial infarction size was significantly decreased in group IP and group DP(P<0.05).But the difference between group IP and group DP was not significant.Compared with group C,there were 8 miRNAs up-regulated and 4 miRNAs down-regulated in group IR;compared with group IP,12 miRNAs were up-regulated and 6 miRNAs were down-regulated in group C;compared with group DP,there were 6 miRNAs up-regulated and 3 miRNAs down-regulated in group C;compared with group DP,there were 16 miRNAs up-regulated and 4 miRNAs down-regulated in group IP;compared with group DP,there were 27 miRNAs up-regulated and 38 miRNAs down-regulated in group IR;compared with group IP,there were 42 miRNAs up-regulated and 72 miRNAs down-regulated in group IR.Conclusions:Dexmedetomidine can play an important role in against myocardial ischemia reperfusion injury;while the mechanism underlying the effect is associated with the miRNAs differential expression.
