Expression And Clinical Significance Of MiR-193a-5p,139-5p,551b-3p,4778-5p In Plasma Of Xuanwei Lung Cancer

Objective:According to the preliminary work and reports,this study mainly to detect the relative expression of miR-193a-5p,miR-139-5p,miR-4778-5p and miR-551b-3p in plasma samples,which from lung cancer patients,Xuanwei lung cancer patients,benign pulmonary disease and healthy physical examination,and to analysis the correlation of clinical data.Then,we explored the value of plasma miRNAs in the diagnosis of lung cancer and Xuanwei lung cancer.Methods:Collected peripheral blood samples from January 2012 to December 2016,and our study contained 51 patients with benign pulmonary disease,53 patients with lung cancer,50 patients with lung cancer in Xuanwei,and 53 normal control peripheral blood samples.The relative expression of miR-193a-5p,miR-139-5p,miR-4778-5p and miR-55 1b-3p in the plasma of the four groups were detected by RT-qPCR.Relative expression of plasma miRNAs between the two groups were compared by Mann-Whitney U rank test,and in multiple groups analyzed by Kruskal-Wallis test.Then ROC curves were established to evaluate the diagnostic value of these four miRNAs in patients with lung cancer or Xuanwei lung cancer.Results:There was no significant difference in relative expression level of miR-193a-5p,miR-139-5p,miR-4778-5p and miR-55 1b-3p in healthy and benign lung disease groups.The healthy group as the normal control group,in lung cancer group,relative expression of miR-193a-5p,miR-139-5p,miR-4778-5p,miR-55 1b-3p were down-regulated(P<0.05);In Xuanwei lung cancer group,relative expression of miR-55 1b-3p was down-regulated(P<0.05),and there was a downward trend of miR-139-5p,miR-193a-5p,miR-4778-5p(P>0.05).The benign lung disease group as the normal control group,in lung cancer group,relative expression of miR-193a-5p,miR-139-5p,miR-4778-5p were down-regulated(P<0.05),and there was a downward trend of miR-55 1b-3p(P>0.05);In Xuanwei lung cancer group,relative expression of miR-193a-5p,miR-4778-5p,miR-551b-3p were down-regulated(P<0.05),and there was a downward trend of miR-139-5p(P>0.05).In lung cancer group,the relative expression level of miR-139-5p was correlated with smoking(P<0.05),and miR-551b-3p was related to lymph node metastasis(P<0.05).The relative expression level of miR-4778-5p was correlated with tumor stage(P<0.05).In Xuanwei lung cancer group,the relative expression level of miR-551b-3p and miR-139-5p was correlated with smoking(P<0.05).The relative expression of miR-193a-5p,miR-139-5p,miR-4778-5p and miR-551b-3p were detected in 42 lung cancer patients and 47 healthy subjects.The ROC curve was established by analyzing the lung cancer group,and area under the ROC curve were:0.672?0.681?0.745 ? 0.623,respectively.The relative expression of miR-193a-5p,miR-139-5p,miR-4778-5p and miR-551b-3p were detected in 42 lung cancer patients and 40 patients with benign lung cancer.The ROC curve was established in lung cancer group,and AUC were 0.807?0.919?0.568 ? 0.893,respectively.The relative expression of miR-193a-5p,miR-139-5p,miR-4778-5p and miR-551b-3p were detected in 43 patients with Xuanwei lung cancer and 47 healthy subjects.The ROC curve was established in Xuanwei lung cancer,and AUC were 0.582,0.555,0.600 and 0.361,respectively.The relative expression of miR-193a-5p,miR-139-5p,miR-4778-5p and miR-551b-3p were detected in 43 patients with Xuanwei lung cancer and 40 patients with benign pulmonary disease.The ROC curve was established in Xuanwei lung cancer,and AUC were 0.519,0.512,0.427 and 0.365,respectively.Conclusion:In group of lung cancer or Xuanwei lung cancer,relative expression of miR-193a-5p,miR-139-5p,miR-4778-5p,miR-551b-3p were down-regulated,and they have a good value in the diagnosis of lung cancer,and their research prospect is worth looking forward to.
