| Objectives:1.To establish a mice model of thyroid cancer suitable for microwave ablation.2.To examine different power seting of microwave ablation through a variety of detection methods to KMH-2 rat model.After treatment,tissue sample were taken to examine pathological changes in tissue by MWA after HE and Tnuel sating.Ssearch for the lowest parameters that cause apoptosis in tumor cells and evaluate the short-term effects of it on KMH-2 thyroid cancer.Methods:Rude mice were randomly divided into 4 group(n=10)and subcutaneously injected with KMH-2 cells of different number(5×105,1×106,2×106 and 4×106)respectively,then the tumor formation rate in the 4 group were compared.The ideal cell number was selected to establish the mice model once again..When the tumor diameter reached 0.8-1.Ocm,the model mice were randomly divided into 7 groups(n=5 in each group)to undergo MWA.The temperature and ablation time was 45℃and 2 minute.Power set group A for the control group which was 0W(n=5),group B was 5W(n=5),group C was 5W(n=5),group D was 5W(n=5),group E was 5W(n=5),group F was 5W(n=5),group G was 5W(n=5).The tissue sample were taken to examine pathological change in tumor after HE and Tunel staining.Observed the imaging changes before and after treatment by using the high frequency color Doppler ultrasonography and elastography and killed one nude mouse at random in each group.Then the tumor model after treatment was fixed and had the HE staining and TUNEL staining to detect the apoptotic index.Analyzed the apoptotic index and elastography results.After treatment,group A(OW),group B(5W),group C(10W),group D(15W),group E(20W),group F(25W),group G(30W),control group,animals were observed for 1 and 30 days,two-dimensional ultrasound examination and elastography were performed.Dissecting the tumor for pathological examination,the apoptosis of each group was detected by HE staining and TUNEL.Observed the survival rate,mental status and recurrence or metastasis of the animals in each group after 1 months of the treatment.Data AnalysisSPSS 19 statistical software was used to analyze the experimental data.Each experiments were repeated 3 times independently.The measurement data was expressed by x±s and the compare of the 2 independent samples adopted the Student t test.The mean between groups was analyzed by variance analysis and LSD-t test was used in pairwise comparison within the group.p<0.05 means that the difference has statistically significant.Results:1.The group with 2×106KMH-2 cell injection had high formation rate and was suitable for MWA.2.A to F group increased with the power,the elastic imaging and measurement of the Young’s modulus of the microstructure were all manifested as hardness,and the apoptosis index of A-D group increased with the increase of power,the difference between each group is statistically significant(P<0.05),when the ablation power rises to the 15W elastic imaging of Young’s modulus reaches 63.2 ± 6.2,the cell apoptosis index reaches the platform stage 0.61 ± 0.16,e group Young’s modulus is 87.6 ± 8.6 Apoptosis index is 0.60±0.15,F Group Young’s modulus is 149.6 ± 22.1,the apoptotic index is 0.61 ± 0.17,C,D,E power the higher the carbonation becomes apparent,the hardness is higher,the three groups of apoptotic indices were not statistically significant(P<0.05)the G-Group apoptosis index dropped to 0.59 ±0.12,Young’s modulus of 149.4 ± 20.1.Conclusion:45℃、2min 15W microwave ablation is feasible and effective for KMH-2 cells subcutaneously implanted model of thyroid cancer mice. |
