Objectives:In this study,the specific IncRNA which may be regulated by Hintl gene in hepatocellular carcinoma cells was screened by high-throughput IncRNA chip.The possible target or signal pathway and molecular mechanism of the IncRNA was explored in vitro.This study provides a preliminary basis for further study of its feasibility as a marker of liver cancer prognosis and a feasibility treatment target.Methods:1.Hintl gene knockdown HepG2 cells were obtained by siRNA interference.The effects of Hintl gene on the proliferation,invasion and metastasis of HepG2 cells were confirmed by MTT,Transwell and scratches;2.The changes of IncRNA in HepG2 cells were detected by high-throughput IncRNA microarray,and the specific IncRNA(IncURHI)was screened by bioinformatics;3.The potential target microRNA(miR-455-5p)of IncURHI was predicted by bioinformatics;4.LncURHI knockdown HepG2 cells were obtained by smart silencer technology and the changes of intracellular miR-455-5p abundance were detected by RT-qPCR;5.The effects of cell proliferation,invasion and metastasis of Hint(-)IncURHI(-)HepG2 cells were observed by MTT,Transwell and scratches.Results:1.The knockdown of Hintl significantly reduced the proliferation,invasion and metastasis of HepG2 cells;2.The specific expression of IncURHI in HepG2 cells knocked down by Hintl was reduced;3.Bioinformatics prediction suggests that miR-455-5p and IncURHI have potential MRES;4.LncURHI knockdown can significantly increase the abundance of intracellular miR-455-5p,but cant reach the level of normal HepG2 cells;5.The proliferation,invasion and metastasis ability of Hint 1(-)IncURHI(-)HepG2 cells were lower than Hint 1(-)cells,but higher than normal HepG2 cells.Conclusions:1.In HCC cells,IncURHI may "sponge" the intracellular miR-455-5p by MREs,exert the characteristics of ceRNA,regulate the abundance of intracellular miR-455-5p,and influence the proliferation,invasion and metastasis of HCC cells;2.It may be one of the mechanisms of Hintl play as a tumor suppressor gene that Hintl can regulate cell proliferation,invasion and metastasis ability by regulating the expression of IncURHI;3.The expression of IncURHI enhances HCC cell proliferation,invasion and metastasis.
