ObjectiveTo investigate the effect and mechanism of lncRNA-AF113014 in the development of HCC.Methods1.lncRNAs differentially expressed in normal hepatocytes and hepatoma cells were verified by q RT-PCR.We chose lncRNA-AF113014 which was down-regulated in HCC cells for our reasch.2.The proliferation of lncRNA-AF113014 on SMMC-7721 and SK-Hep1 cells were examined by MTS and clone formation assay.3.Found the target gene of lncRNA-AF113014.qRT-PCR and Western blot analysis were used to testify the expression of target gene in SMMC-7721 cells.4.The biological functions of the target gene Egr2 were detected by MTS and clone formation assay.5.The relationship of lncRNA-AF113014,mi RNA-20 a and Egr2 wasanalyzed by luciferase assay.6.Tumor formation in nude mice,immunohistochemistry and Western blot were used to testify the effect of lncRNA-AF113014 on tumor growth and Egr2 in vivo.Results1.lncRNA-AF113014 were down-regulated in HCC cells.2.lncRNA-AF113014 inhibited the proliferation of hepatoma cells.3.lncRNA-AF113014 promoted the expression of target gene Egr2.4.Egr2 suppressed the HCC proliferation.5.lncRNA-AF113014 promoted the expression of Egr2 by interacting with miRNA-20 a.6.lncRNA-AF113014 inhibited tumor growth and promoted Egr2 expression in vivo.ConclusionlncRNA-AF113014 can promote the expression of Egr2 by interacting with miRNA-20 a and inhibit the the proliferation of hepatoma cells. |
