Objective:the present treatment of large area burn patients still exist certain problems,how to as soon as possible in the early stages of a closed wound,make the patients as soon as possible into the plateau,is the clinical doctors and researchers have been trying to solve clinical problems.In recent years on Mesenchymal Stem Cells(Mesenchymal Stem Cells,MSCs)research in an endless stream,the researchers on the characteristics of Mesenchymal Stem Cells are very interested in between,especially between recently Umbilical Cord Mesenchymal Stem Cells,Umbilical Cord Mesenchymal Stem Cells,UCMSCs)is to show its advantages,such as extraction,convenient and self-renewal ability and so on,it also makes many researchers interested in it,to study the tireless exploration.In this study,the repair ability of the filling stem cells in the umbilical cord of the umbilical cord was investigated.Model experiment first,according to burn out ideal burn model preparation method,and then according to this method for preparation of burn model,using the method of on-spot application of umbilical cord mesenchymal stem cells for the treatment of wounds in rats,to explore the umbilical cord mesenchymal stem cells to burn wounds between regulation,and its effects on vascular endothelial growth factor regulation,for clinical treatment by studying the exploration can burn patients,repair provides some new ideas and ways of the woundMethods 1:Three methods of preparation of burn burn model for SD rats were adopted by three methods(burn method,hot solid burn method,hot water burn method),and the differences of three methods were compared.Burn method is used for barbecue flamethrower to burn rats back,hot solid burn method for water pot of heated copper bar in the back to scald rats,hot water scald method for special centrifugal tube placed in rats back,and then into the centrifuge tube in a fixed quantity of hot water,hot water and the rat skin contact after fixed time back skin burns in rats.After burning and burning the rat,the rat had a normal daily diet and observed the basic condition of the rats and the changes in the surface.Burn in rats after 3 days to remove back injury rats skin,paraformaldehyde fixed preservation,sucrose solution dehydration,skin tissue for frozen section again after HE staining,observed the pathological situation of each rat skin tissue injured.Results 1:Three approaches to the back skin of SD rats were skin lesions caused by certain extent,but the comprehensive all aspects to consider,the third group of hot water scald burn area and depth of the controllable method,the required equipment is easy to get,by contrast is more suitable for repetitive operations.It can be used for the preparation of small animals such as SD rats,rabbits,and naked mice.Conclusion 1:three methods have a certain degree of skin injury of back skin of SD rats,but all aspects of comprehensive consideration,third methods of hot water scald group is the most satisfactory,compared with more suitable for repetitive operation.It can be used for the preparation of small animal models such as SD rats,rabbits and nude mice.Methods 2:The preparation of the back burn model for 30 SD rats was applied by hot water scalding,and the rats were randomly divided into 3 groups of 10.In rats with scald after removal of a skin scald rats back on the third day,measure the size of the wound after resection,photographed after wound size was calculated by the image analysis software ipp6(the original area of rat wound area).And given different treatment according to different groups,the treatment of the first group of 1 ml concentration(1 x106/ml)of umbilical cord mesenchymal stem cells between +autologous skin treatment,the second group of treatments for autologous skin treatment,the third group as the blank control group,not on medication,rat wound last three group were applied with covering the wound,gauze packing fixed.Situation of rats after daily observation,every 7 days after the treatment of wounds in rats,after statistical analysis,the healing area is calculated to evaluate wound healing in rats.The skin was taken from the skin after the healing of the rat,and the skin tissue was observed and the immunofluorescence checked VEGF.Results2:The three groups of rats were successful in applying hot water scalding,and if they were done properly,they were less likely to be different.Through the study of the statistical evaluation of rat wound healing area,according to the results in 2,3,4,5,and 6 weeks the first set of wound healing faster than the second group,the first group and the third group wound healing without statistical significance;In the second,third and fourth weeks,the third group healed faster than the second group,and the second and third groups healed at no statistical significance in the fifth and sixth weeks.The first group of the three groups was the most common in the third group of rats.The first group of slices showed the expression of multiple capillaries,the highest amount of collagen in three groups,and the collagen percolation.The second group of rats showed that the collagen permutations in the tissues were less than the levels of the first group of rats.This experiment immunofluorescence test result shows that the first set of umbilical cord mesenchymal stem cells and between autogreffe DE la peau treatment group the expression of vascular endothelial growth factor is higher than the second group of separate autologous skin treatment group and the third group of the blank control group.Conclusion 2:The filling stem cells of the umbilical cord can accelerate the speed at which the rat's skin can.repair burn wounds.Secondary umbilical stem cells may accelerate the healing of the wound by promoting the proliferation of VEGF.
