Chinese herbal medicine is a complicated multicomponent system.The quality control of medicinal materials has a significant meaning to ensure clinical efficacy,modernization of traditional Chinese medicine and promote the Chinese medicine's industry.Therefore,a simple and convenient method of quality analysis is necessary for content determination precisely and quickly.The technology of near infrared spectroscopy(NIR)is a rapid and non-destructive environmental analysis method combined with stoichiometric means to determine medicinal materials' content rapidly.This research uses it for reference to get the purpose of content determination for Paeoniae Radix Rubra,Astragali Radix,Fructus Gardeniae,Rhei Radix et Rhizoma,Angelicae Pubescentis Radix,Saposhnikoviae Radix,Angelica Dahurica Radix.Progress made as the following:1.High performance liquid chromatography(HPLC)was applied to determine the content of gallic acid,catechin,albiflorin and paeoniflorin in Paeoniae Radix Rubra;101 samples were chose,build a correction model between their content and near infrared spectroscopy by partial least squares(PLS).Result displayed as follows:by the method of multiplicative scatter correction and 2nd derivative,the correlation coefficient of gallic acid is 0.9065,the value of root mean square error of calibration(RMSEC)is 0.10,the validation set of correlation coefficient is 0.9163,the value of root mean square error of prediction(RMSEP)is 0.09.By the method of multiplicative scatter correction and 2nd derivative,the correlation coefficient of catechin is 0.9408,the value of RMSEC is 0.44,the validation set of correlation coefficient is 0.9231,the value of RMSEP is 0.40.By the method of multiplicative scatter correction and 1st derivative,the correlation coefficient of albiflorin is 0.9406,the value of RMSEC is 0.13,the validation set of correlation coefficient is 0.9226,the value of RMSEP is 0.14.By the method of Savitzky-Golay filter and 1st derivative,the correlation coefficient of paeoniflorin is 0.9674,the value of RMSEC is 0.22,the validation set of correlation coefficient is 0.9281,the value of RMSEP is 0.22.It has a nice linear relationship between the measured values of HPLC and NIR model.This means that the model has such a good fitting ability as to determine the gallic acid,catechin,albiflorin,paeoniflorin's content of Paeoniae Radix Rubra.2.High Performance Liquid Chromatography coupled with Mass Spectrometry is applied to determine the content of Calycosin-7-glucoside and astragaloside in Astragali Radix.91 samples were chose,build a correction model between their content and near infrared spectroscopy by PLS.Result displayed as follows:by the method of multiplicative scatter correction,1st derivative and Savitzky-Golay filter,the correlation coefficient of Calycosin-7-glucoside is 0.8635,the value of RMSEC is 0.019,the validation set of correlation coefficient is 0.8266,the value of RMSEP is 0.023.By the method of 2nd derivative and Savitzky-Golay filter,the correlation coefficient of astragaloside is 0.7963,the value of RMSEC is 0.008,the validation set of correlation coefficient is 0.8548,the value of RMSEP is 0.006.It has a nice linear relationship between the measured values of HPLC and NIR model.This means that the model has such a good fitting ability as to determine the Calycosin-7-glucoside and astragaloside's content of Astragali Radix.3.Quantitative analyzed to geniposide by mid-infrared spectroscopy(IR)and near-infrared spectroscopy(NIR).First,according to Pharmacopoeia method,100 samples were chose,build a correction model between their content and NIR.IR spectroscopy by PLS.Result displayed as follows:by the method of 2st derivative and Savitzky-Golay filter,the correlation coefficient of geniposide is 0.9725,the value of RMSEC is 0.20,the validation set of correlation coefficient is 0.9606,the value of RMSEP is 0.22 in NIR.By the method of 2nd derivative and standard normal variate(SNV),the correlation coefficient of geniposide is 0.9256,the value of RMSEC is 0.24,the validation set of correlation coefficient is 0.9174,the value of RMSEP is 0.24.It is showed by the experiment that IR spectroscopy and MIR spectroscopy both can determine the geniposide of Fructus Gardeniae.4.Use pharmacopoeia method for reference and NIR spectroscopy to determine the content and the total content of emodin,chrysophanol,rhein,aloe-emodin,physcion in Rhei Radix et Rhizoma from 106 samples;and build a best correction model between it and near infrared spectroscopy through an inspection of pretreatment method.Result displayed:by the method of 1st derivative,the correlation coefficient of mdoel is 0.9856,the value of RMSEC is 0.13,the validation set of correlation coefficient is 0.9445,the value of RMSEP is 0.23.It is showed by the experiment that NIR spectroscopy can determine the total content of emodin,chrysophanol,rhein,aloe-emodin,physcion in Rhei Radix et Rhizoma.5.Use pharmacopoeia method for reference and NIR spectroscopy to determine the content of osthole and columbianadin in Angelicae Pubescentis Radix from 97 samples;and build a best correction model between it and near infrared spectroscopy through an inspection of pretreatment method.Result displayed:by the method of 1st derivative,the correlation coefficient of osthole is 0.9233,the value of RMSEC is 0.16,the validation set of correlation coefficient is 0.9413,the value of RMSEP is 0.14.By no method of pretreatment,the correlation coefficient of columbianadin is 0.8315,the value of RMSEC is 0.11,the validation set of correlation coefficient is 0.8574,the value of RMSEP is 0.10.It is showed by the experiment that NIR spectroscopy can determine the content of osthole and columbianadin in Angelicae Pubescentis Radix.6.Use pharmacopoeia method for reference and NIR spectroscopy to determine the content of imperatorin in Saposhnikoviae Radix from 99 samples;and build a best correction model between it and near infrared spectroscopy through an inspection of pretreatment method.Result displayed:by the method of 2nd derivative and Savitzky-Golay filter,the correlation coefficient of imperatorin is 0.9755,the value of RMSEC is 0.01,the validation set of correlation coefficient is 0.9630,the value of RMSEP is 0.01.It is showed by the experiment that NIR spectroscopy can determine the content of imperatorin in Saposhnikoviae Radix.7.Use pharmacopoeia method for reference and NIR spectroscopy to determine the content and the total content of prim-O-glucosylcimifugin and 5-0-methylvisammioside in Angelica Dahurica Radix from 100 samples;and build a best correction model between it and near infrared spectroscopy through an inspection of pretreatment method.Result displayed:by the method of 2nd derivative and MSC,the correlation coefficient of model is 0.9773,the value of RMSEC is 0.05,the validation set of correlation coefficient is 0.9617,the value of RMSEP is 0.051.It is showed by the experiment that NIR spectroscopy can determine the total content of prim-O-glucosylcimifugin and 5-O-methylvisammioside in Angelica Dahurica Radix.
