Objective: This study aims to determine the expression of imprinting genes Grb10 and H19 in skeletal muscle after acute injury and to explore the relationship between imprinting genes Grb10 and H19 with the process of skeletal muscle repair after injury.Participants and Methods: 30 adult male SD rats;anesthetizing with pentobarbital sodium;intraperitoneal injection of 50 mg/kg each rat.After anesthesia was conducted,the acute blunt trauma model of rat skeletal muscle of Kam's and Chen shiyi's as reference were used.The rats of skeletal muscle acute injury model were randomly divided into 5 groups with the computer software random number method.Meanwhile,8 rats will be put in each group.It is necessary to draw the materials after duplicating the mold 2nd days,4th days?6th days?8th days ?10th days.Marked the tissue of middle in the left side of the skeletal muscle of rats as the experimental group,for the normal right side as the control group.By the way of serving the morphologic changes of the acute injury skeletal muscle with the HE staining.Real-time RT-PCR and western blotting were used to assess the expression level of Grb10 and H19.Results: HE staining pictures result display that the skeletal muscle fibers aligned disorderly.From that,we can find that at the 2nd day,the nucleus number began to increase,and the nucleus number much more than the the nucleus number of the control group significantly at the 4th day,at the 6th 8th 10 th,the nucleus number significantly declined.Meanwhile,there is no significance exists with the control group.From the result of Real-Time PCR,Grb10 and H19 expressed both in the experimental group and the control group.2nd the expression of Grb10 was increased,4th the expression level increased significantly,its expression was twice over the control group,with significant difference,6th declined obviously,and the 8th and 10 th the expression level is stable;2nd the expression of H19 was decreased,4th the expression level increased significantly,its expression with significant difference,6th declined obviously,and the 8th and 10 th the expression level is stable.Grb10 western blotting results show that: the expression level of Grb10 protein expression in skeletal muscle injury after 2 days has increased significantly and get to the maximum at 4th,from 6th to 10 th,Grb10's expression level was significantly declined,even lower than the expression level of the control group.Conclusion: 1)The expression level of imprinting genes Grb10 and H19 had significant difference in the repair process of skeletal muscle after acute injury,so we infer that the imprinting genes Grb10 and H19 did influenced the repair of skeletal muscle after acute injury.2)It is presented that the expression level of imprinting genes Grb10,which is similar to the expression level of imprinting genes H19.Under this circumstance,the conclusion could be inferred that there may be some relationship between the imprinting genes Grb10 and H19 during the repair process of skeletal muscle after acute injury.
