Objective: To explore wether the ginkgo biloba extract play a part in intermittent hypoxic rat thoracic aortic reconstruction.Methods: Choosing 30 healthy male wistar rats divided into 3 groups(N=10)randomly: normal control group(NC),chronic intermittent hypoxiagroup(CIH),chronic intermittent hypoxia with GBE intragastric administrationgroup(CIH+GBE).Checking the pathological changes about thoracic artery by hematoxylin(HE)staining.mRNA expression of ERK1/2 ? p38 MAPK ? JNK genes were analyzed by qRT-PCR.Detecting the protein level of such factors above and p-ERK1/2 by immunoblotting.Texting the level of TNF-? ?hs-CRP and IL-6 in serum by ELISA.Results:(1)According to hematoxylin(HE)?Masson staining vasculature remodeling was happening in the CIH group accompanying with thoracic artery wall thickening,increased fibrosis and accumulation of extracellular matrix comparing to the normal(P<0.05).However,when compares CIH to CIH+GBE,the results come out to be different(P<0.05).(2)Accordingto NC group,Expression analysis of JNK?p38MAPK by qRT-PCR significantly increased in thoracic arteryof the CIH group(P<0.05).While the p38MAPK?JNK was significantly decreased in CIH+GBE,according to the CIH(P<0.05).The expression level of ERK1/2 protein and its expression of mRNA in all group were similar to the nomal.(3)The protein expression of p38MAPK?JNK?ERK1/2 was identify with its expression level of mRNA,but not the same with p-ERK1/2.Compared with NC,the protein expression of p-ERK1/2 in CIH was significantly increased,when CIH+GBE compares to CIH,the protein expression of p-ERK1/2 in CIH+GBE was significantly decreased(P<0.05).(4)ELISA showed that: The levels of TNF-?,hs-CRP and IL-6 in CHI were significant higher than those in NC(P<0.05),and the content of TNF-??hsCRP was lower in GBE compared with CIH(P<0.05).While IL-6 made no diference.Conclusion: Intermittent hypoxia may result in abnormal inflammatory reactionand and thoracic artery remodeling.GBE can be expected to be an hopeful drug to treat and prevent the artery remodeling caused by OSAS. |
