Objective:Endometriosis(EMs)is a benign disease,which shows similar biological behaviors to malignant tumors,such as infiltrating and damaging surrounding tissues,distant metastasis and recrudescing easily,therefore,the pathogenesis of EMs may be some things in common with malignant tumors.Last few years,eutopic endometrium of EMs as the objectives were studied by researchers around the world.These studies indicate that EMs is a polygenic and multiple factors related disease,which differs in the expression of genes and proteins compared with normal endometrium.Researchers identified 10 up-regulated genes in the eutopic endometrium of endometriosis during the secretory phase using cDNA-RDA and found that ATPase family AAA domain containing 3A(ATAD3A)was one of the up-regulated genes,which has closed relation to biological characteristics,such as cell proliferation,anti-apoptosis,anti-autophagy,infiltration,metastasis and drug-resistance and so on.In this study,we study ATAD3 A expression of the endometriosis(EMs)in the level of mRNA and protein,and explore the deep connection between ATAD3 A and EMs.Materials:All the tissues were adopted from patients in the first affiliated hospital of NanChang university from March to August in 2016.Ectopic endometrial tissues(n=31)were obtained from these women who sufferred from the ovarian cyst removal surgeries,and were diagnosised histopathologically as endometriosis.Their average age was 42±3.27.Eutopic endometrial tissues(n=31)were obtained from those women which diagnosed as endometriosis during surgery.Their average age was 42±3.27.Normal endometrial tissues(n=31)were obtained from these women who were normal,and their average age was 41±4.16.All these participants without physical diseases have a regular menstruation,about 28-32 d,and they hadn’t accepted hormones,other drugs or intrauterine devices within the three months before surgery.All collected tissues were cleaned by PBS,ones put into liquid nitrigen for extracting mRNA and protein,the others were fixed by stationary liquid forimmunohistochemistry.Methods:A quantitative analysis of the expression of ATAD3 A mRNA and protein in the endometrium was detected by Quantitative real-time polymerase chain reaction(qPCR),western blot and immunohistochemistry in patients with endometriosis(ectopic group,n=31;eutopic group,n=31)and patients with normal endometrium(control group,n=31).These experimental results were analysised by 3 groups homogeneity of variance analysis and single factor variance analysis calculated by SPSS19.0,with P<0.05 for the difference was statistically significant.Results:1.The expression of ATAD3 A mRNA were significantly higher in ectopic(3.04±0.73)and eutopic(2.30±0.83)endometrium than in normal(1.12±0.48)endometrium(P<0.05),there was statistcal difference between ectopic and eutopic endometrium(P<0.05).2.The expression of ATAD3 A protein were significantly higher in ectopic and eutopic endometrium than in normal endometrium(P<0.05)。There was statistcal difference between ectopic and eutopic endometrium(P<0.05).Conclusions:There is expression of ATAD3 A m RNA and protein among these group.Both the expression of ATAD3 A mRNA and protein were significantly higher in ectopic and eutopic endometrium than in normal endometrium,it indicates that ATAD3 A may be a significant genetic target which can induce endometrial cell to implant at abnormal sites,the correlation between ATAD3 A and EMS needs to be further verified. |
