Objective:Detect 5-Fu pharmacokinetic parameters of advanced colorectal cancer patients receiving FOLFOX6 [oxaliplatin +calcium folinate+ 5-fluorouracil(5-Fu)].To analyze the correlation of AUC in reducing the incidence of adverse reactions and improving the therapeutic effect.Methods:Forty patients with advanced colorectal cancer treated with FOLFOX 6regimen were selected.Based on body surface area administration,receiving two rounds of chemotherapy.The patients were randomly divided into the control group(n = 20)and the experimental group(n=20).The first cycle of chemotherapy in all patients were the traditional body surface area(BSA)mode of administration,the second cycle to the end of chemotherapy,the control group is still using BSA administration.The concentration in experim ental group of 5-Fu in peripheral blood was measured by 5-Fu intravenous infusion for 18-30 h in the first cycle of chemotherapy,and the dose of 5-Fu was adjusted according to the pharmacological parameters of 5-Fu.After the two rounds of chemotherapy,the adverse reactions and efficacy of the patients were analyzed.Results:The incidence of diarrhea in I~II lever,gastrointestinal reaction and mucositis(30%,25%,25%)in the experimental group was lower than that in the control group(65%,60%,65%)(P <0.05).The incidence of myelosuppression and gastrointestinal reaction in III ~ IV lever about the experimental group(10%,10%)was lower than that in control group(45%,40%)(P <0.05).The overall response rate(60%)in the experimental group was higher than that in the control group(25%)(P <0.05).Conclusion:Real-time monitoring 5-Fu blood concentration with FOLFOX6 chemotherapy in patients of advanced colorectal cancer,and adjust the dose according to pharmacokinetic parameters AUC,will effectively reduce the toxicity and improve the therapeutic effect.
