Objectives: Detection of the peripheral blood of the change of survivin/caspase3, CD44V6of the colorectal cancer patients with stage (Dukes B, C) surgery after intraoperative implantation of fluorouracil implants, discusses its inhibitory effect on human colorectal cancer, micro metastasis and prognosis of tumor and the effect of preliminary from clinical trials to clarify fluorouracil implants may play induced apoptosis mechanism, for the clinical application of fluorouracil implants antitumor provides a more direct and more reliable experimental basis.Methods:64patients with colorectal cancer (stage Dukes B, C) according to random number table method is divided into the treatment group and control group,32cases in each group. Standard treatment group underwent radical surgery, intraoperative implantation of fluorouracil implants; control group given only standard radical surgery and surgery is not applied fluorouracil implants.1. Semi‐quantitative RT‐PCR method was measured before and after surgery in patients with colorectal cancer in peripheral blood of survivin, caspase‐3expression levels.2. Using flow cytometry test peripheral blood in patients with colorectal cancer before and after operation of CD44V6content.Results:1. All patients in the preoperative and postoperative1d to14d, early in the morning on an empty stomach EDTA anticoagulant tube venous sampling2mL, in which patients with preoperative1d peripheral blood test results show that: Semi‐quantitative RT‐PCR assay in the control group before surgery survivin was (1.090±0.1863), caspase‐3was (1.134±0.0082), and flow cytometry to detect CD44V6is (9.729+0.943), same as above method,the preoperative treatment group survivin was (1.157±0.1662), caspase‐3was (1.119±0.0295), CD44V6was (9.557±1.170), the two groups of preoperative each factor level difference is no statistical significance (P>0.05).2. Semi‐quantitative RT‐PCR method for detection of two groups of patients with postoperative peripheral blood survivin/caspase3expression level, the results showed that the control of postoperative survivin was (0.585±0.207), caspase‐3was (1.654±0.111), postoperative survivin as treatment group (0.362+/‐0.183), caspase‐3(2.001+0.146), it is obvious that the expression of survivin levels lower than the control group after treatment group, treatment group of caspase3after the expression level is higher than the control group, two groups of postoperative survivin/caspase3level difference was statistically significant (P <0.05).3. Flow cytometry method assay the expression level of CD44V6in two groups of patients with postoperative peripheral blood,the results showed that the control of postoperative CD44V6was (3.471+0.496), postoperative CD44V6as treatment group was (1.857+0.535), it is obvious that the treatment group of postoperative CD44V6expression level significantly lower than the control group, two groups of postoperative CD44V6level difference was statistically significant (P <0.05).Conclusion: Fluorouracil in patients with colorectal cancer by sustained release implant of peripheral blood laboratory tests, which can be found via downregulation of survivin, caspase‐3expression is up‐regulated manner to promote apoptosis, which may be fluorouracil implants one of the mechanisms to promote apoptosis in colorectal cancer. Its mechanism of action in terms of tumor cell apoptosis, for the treatment of colorectal cancer provides new ideas. By down‐regulating its expression of survivin, inhibits tumor angiogenesis, and thus play its inhibition of tumor growth and metastasis. The reduction of the expression of its CD44V6may inhibit a variety of tumor cell adhesion between residual disease kill to achieve resection surgery is not micro‐metastases and intraperitoneal free cancer cells, which effectively kill micro‐metastases, tumor prognosis and recurrence of benign effects may exist.
