Objective: This study aimed to investigate the relationship between single-nucleotide polymorphisms of myeloperoxidase(MPO)and Leukoaraiosis(LA)in patients with acute ischemic stroke.Methods: 470 cases of acute ischemic stroke admitted to the neurological department of Yijishan Hospital between January 2014 and March 2016 were enrolled into our study that were divided into LA group and non-LA group.The correlation between plasma MPO and LA was assessed by enzyme linked immunosorbent assay.Genotyping was performed using polymerase chain reaction and high-resolution melt analysis for the two hypothesized SNPs.Genotypes and allele frequencies of cases and controls were evaluated.Odds ratios were calculated for allele frequencies.Results:(1)The study indicated that plasma MPO was associated with LA and positively correlated with the severity of LA;(2)For rs2243828,we found that LA was significantly associated with the GG genotype as compared with AA(X~2 =6.948,P=0.010,OR =0.388,95%CI =0.188~0.799).Minor allele(G)of rs2243828 was found to be significantly associated with LA(X~2=17.722,P=0.000,OR=0.527,95%CI=0.390~0.712).Regarding rs2107545,no significant difference in the genotypes and allele frequencies were found between LA patients and controls.Conclusion: Our results suggested MPO was a risk factor for LA.Our results also suggested that rs2243828(A/G)of MPO gene were significantly associated with LA,G allele played a protective role in the presence of LA.This effect may be mediated by the effect of this polymorphism on the transcription of MPO gene.
