Objective: To detect the expression of HEATR1(HEATR repeat containing 1)in two strains of gastric cancer cells BGC-823 and AGS,when its overexpressed in gastric cancer cells;and detect the positive expression of HEATR1 In gastric cancer tissues and normal gastric tissue;To observe the normal gastric mucosa HEATR1 in paraffin section and gastric cancer tissue paraffin section gene expression.Method: Primer sequences swas designed and synthesized by Gene synthesis company according to the nucleotide sequence of HEATR1 m RNA reported in Gen Bank.Gastric cancer cells after vaccination in six holes culture plate,Using the PCR detect endogenous gene expression in gastric cancer cell line BGC-823 and AGS,when confirmed its high expression in gastric cancer cells,Q-PCR detection HEATR-1 m RNA expression level between the preoperative diagnosis and surgery of gastric cancer patients with gastric cancer tissue line and normal tissue line,using immunohistochemical method evaluate HEATR-1 in the paraffin section of the gastric cancer,and using immunohistochemical grading method,exploring the relationship of HEATR-1 protein positive expression and histological types of gastric cancer.Results:The HEATR1 gene overexpressed in two strains AGS and BGC-823 gastric cancer cells(AGS ? Ct average = 7.10,BGC-823 average ? Ct = 6.59,relative expression m RNA were7.10±0.111,6.59±0.123)in the gastric cancer cell lines endogenous gene detection.Line 20 cases of gastric cancer patients with gastric cancer tissue and negative cut edge line Q-PCR detection HEATR-1 m RNA expression capacity,high expression of 12 patients,8 cases of lower expression,the relative contents of m RNA line matching t test between the two groups,P < 0.05,the difference was statistically significant.Selection of gastric cancer and normal gastric mucosa paraffin section of the 12 cases respectivly.Gastric cancer tissue paraffin sections of HEATR1protein were significantly higher expressed in normal gastric tissues.HEATR1 protein was mainly localized and overexpressed in the tumor cell cytoplasmand cytolemma.The average IHC score of HEATR1 expression in gastric tissue and normal gastric mucosa tissues was(4.84±1.642)and(1.92±0.900),respectively.Comparison between normal group and gastric cancer group differences in immunohistochemical staining via the unpaired t test,P=0.032,P < 0.05,the difference was statistically significant.All of these results show that HEATR1 high expression may be associated with the occurrence and development of gastric cancer.Conclusion: HEATR-1 gene overexpressed in gastric cancer cell lines and gastric cancer tissue,it could be a disease-causing genes in gastric cancer,and it expected to become a new molecular therapy target of gastric cancer in the future.
