Objective:In this study,SD(Sprague dawley)subarachnoid hemorrhage rat model was used to study the morphological level,molecular level and drug intervention.HE staining,WB and RT-PCR were used to observe the subarachnoid Expression of S1PR1(Sphingosine-1-Phosphate Receptor 1,S1PR1)and TNF-? in Brain Tissue of Rats with Hemorrhage.The effects of edaravone on the expression of S1PR1 and the effects of edaravone on brain injury in the early stage of subarachnoid hemorrhage were studied and discussed.Methods:70 clean grade male healthy SD rats which were 70 ~ 12 weeks old,weighing 250 g ~ 300 g,the sham operation group(n=20),early subarachnoid hemorrhage(n = 20)and drug intervention group(n =20).The early Brain Injury(EBI)animal model was established by intravascular puncture.The rats in the intervention group were treated with edaravone.After 24 hours of successful modeling,the animal behavioral score was used to evaluate the neurological deficits of SD rats,and the scores of neurological deficits in each group were compared.The pathological changes of the brain tissue were observed by HE staining.The neuronal necrosis of the rats was observed and the degree of braininjury was evaluated.The expression of S1PR1 protein and TNF-? in brain tissue of rats was detected by Western Blot,and the expression of S1PR1 protein in brain tissue of rats was compared.The expression of S1PR1 mRNA in the brain tissue of each group was detected by rt-PCR,and the expression of S1PR1 mRNA in the brain tissue of rats was compared.Results:1.Comparison of neurological deficits in each group: There were no significant differences in neurological deficit between the blank control group and the sham operation group.The scores of neurological deficits in the early brain injury group and the drug intervention group were 0 ~Rats in the 4-point and 0-point rats were excluded and randomized in the same batch of rats.At the end of the experiment,the neurological deficit score of the rats in the intervention group was significantly lower than that in the spider web The difference was statistically significant(P <0.05)in the early stage of brain injury.2.HE staining results: In the blank control group and sham operation group,we can see that the brain structure of SD rats brain structure is complete,the boundary is clear,no interstitial edema,no obvious obstruction.In the early stage of subarachnoid hemorrhage,the neuronal cells in the brain tissue of the rats were necrotic,and the number of the cells was decreased.Compared with the early brain injury group,theintervention group and the subarachnoid hemorrhage group were relatively Neurons,the structure is slightly clearer,the degree of brain damage was significantly reduced.3.Western Blot test results: The expression of S1PR1 in the brain injury group was significantly lower than that in the sham operation group and the normal control group(P <0.05).The expression of S1PR1 in the drug intervention group was significantly higher than that in the sham operation group and the normal control group(P < 0.05).4.Rt-PCR test results: The expression of S1PR1(CD363)-mRNA in the brain injury group was significantly lower than that in the control group(P <0.05).The expression of S1PR1(CD363)-mRNA in the early stage of subarachnoid hemorrhage was significantly different from that in the control group(P <0.05).Conclusions:1.The expression of S1PR1 protein in brain tissue of SD rats with early subarachnoid hemorrhage was decreased.2.Edaravone can promote the expression of S1PR1 protein in brain tissue of SD rats with early subarachnoid hemorrhage,which can relieve the inflammatory response and promote the cerebral protective effect on brain injury in rats with subarachnoid hemorrhage.
