Ribonucleic Acid(RNA)is an important biomacromolecule in cell and is widely involved in the expression and regulation of genes.RNA is mainly composed of 4 ribonucleotides,namely uracil(U),guanine(G),cytosine nucleotide(C),adenine nucleotide(A).These 4 ribonucleotides link with each other through an 3,5-phosphate diester linkages to form an chain of RNA.In cell,RNA is transcripted from DNA.Ribonucleic Acid ? for Injection is RNA extracted from calf pancreas.In clinical practice,Ribonucleic Acid ? for Injection is widely used in the adjuvant treatment of hepatitis B,pancreatic cancer,liver cancer,stomach cancer,lung cancer,breast cancer,soft tissue sarcoma and other cancer.This paper mainly studied the quality inspection methods of Ribonucleic Acid ? for Injection.Developed a method using HPLC to quantify the bases in Ribonucleic Acid ? for Injection by hrdrolysis with perchloric acid.We found that the content of uracil is different in different batches of products.The lower of uracil,the higher of other impurities and also the more of clinical feedback of adverse reactions.This indicate that we may use the uracil content as a quality inspection standard.We investigated the accuracy of UV absorption method for RNA quantification and found that compared with hydrolysis method,UV absorption method is less reliable.Compared with the quantification of DNA content by diphenylamine method and fluorescent dye method,it was found that there was a correlation between DNA and protein content,suggesting that the two were in the form of a combination.
