Objective: The primary hepatocellular carcinoma(HCC)incidence rate has increased year by year in the worid,ranked second in the tumor related death.If primary liver cancer can get early diagnosis,the patient can obtain a relatively good therapeutic effect.then improve the quality of life and prolong the survival time of patients.The main methods in clinical diagnosis of liver cancer were pathological examination,imaging examination and serum alpha fetoprotein(alpha-fetoprotein,AFP)detection etc.Therefore,searching of new and better serum tumor marker for HCC is very important.To investigate the expression level of serum miR-143/145 in liver cancer and the clinical value of the diagnosis of liver cancer.Methods: The serum of meet the requirements of the liver cancer,cirrhosis of the liver in 30 cases and 30 cases of healthy control were included in this study,The expression level of miR-143/145 in liver cancer,liver cirrhosis and healthy persons were detected by real-time fluorescence quantitative PCR technology.The sensitivity and specificity of serum miR-143/145 in the diagnosis of liver cancer were analyzed by receiver operating characteristic(ROC)curve,and determine the best diagnostic threshold.Results:The highest expression level of miR-143/145 in patients with hepatocellular carcinoma,cirrhosis of the liver second,the lowest normal.The difference between the 3 groups was statistically significant(P<0.05).There was no significant difference in the expression of miR-143/145 in different stages of liver cancer(P>0.05).There was no significant correlation between the expression levels of miR-143/145 with ALT,AST and AFP in the serum of HCC patients(P>0.05).The miR-143 diagnosis AUC of hepatocellular carcinoma was0.86,The best threshold value was 1.30,the sensitivity was 0.871,the specificity was 0.733;The miR-145 diagnosis AUC of hepatocellular carcinoma was0.70,The best threshold value was 1.05,the sensitivity was 0.806,the specificity was 0.600.The diagnostic efficacy of miR-143+AFP,miR-145+AFP and miR-143+miR-145+AFP in hepatocellular carcinoma was higher.But miR-143+miR-145+AFP was the most effective one(AUC=0.994).The highest diagnostic sensitivity was miR-143+AFP.Conclusions: miR-143/145 is closely related to the incidence of hepatocellular carcinoma,and it can be used as an effective marker for diagnosis of liver cancer,Combined detection of miR-143,miR-145 and AFP can detect liver cancer more effectively.
