Objective The levels of serum QSOX1,GSN,DKK1 and AFP were detected in PLC group,other liver diseases group and normal control group to explore the significance of QSOX1,GSN,DKK1 in the diagnosis and prognosis of liver cancer.Methods 60 PLC patients,60 patients with benign liver disease and60 cases of healthy physical examination in our hospital were divided into PLC group,other liver diseases group and healthy control group respectively.ELISA was used to quantitatively detect the concentration of serum QSOX1,GSN and DKK1 protein in each group.ECLIA was used to quantitatively detect the concentration of serum AFP protein in each group.Results 1.The expression level of serum QSOX1 in normal control group,other liver diseases group,PLC group were 4.73(3.82~6.54)pg/ml,29.63(6.47~62.08)pg/ml,90.30(33.30~130.66)pg/ml respectively.Serum GSN expression level in the normal control group,other liver diseases group and PLC group were 7.65(5.94~9.56)ng/ml,60.34(27.50~120.66)ng/ml,113.31(55.20~151.97)ng/ml respectively.Serum DKK1 expression level in thenormal control group,other liver diseases group and PLC group were 195.81(39.21~339.19)pg/ml,369.81(248.76~570.99)pg/ml,941.63(293.31~1521.65)pg/ml respectively.PLC group of serum QSOX1,GSN,DKK1 expression levels were significantly higher than those in other liver diseases group and normal control group,the differences were statistically significant(P <0.05).2.The area under the ROC curve of QSOX1,GSN,DKK1,AFP alone in the diagnosis of PLC were 0.891,0.801,0.760,0.802 respectively,the sensitivity were 88.3%,70%,53.5%,73.3% respectively,the specificity were70%,83.3%,98.3%,81.7% respectively,and Youden index were 58.3%,53.3%,51.6%,55.0% respectively.Among them,the Youden index of QSOX1 was higher than GSN,DKK1,AFP,but the difference was not statistically significant(p>0.05).3.Through the establishment of logistic regression model by QSOX1,GSN,DKK1 to diagnosis PLC.The area of under ROC curve was 0.935,and the sensitivity,specificity and Youden index were 83.3%,92.5%,75.8% respectively.Its Youden index was higher than that of QSOX1,GSN,DKK1,AFP,and the difference was statistically significant(p<0.05).4.In this study,60 patients with PLC and their family members were followed up.It was found that there was no significant difference in the expression levels of QSOX1,GSN and DKK1 between the patients with PLC of death group and survival group during 1 years.Independent risk factors of PLC patients that were analysis by Cox regression.The HR(95%CI)of BCLC stage,CA125 concentration,treatment way were 3.748(1.417-9.913),2.571(1.129-5.856),3(1.068-8.428)respectively.Conclusion 1.Serum QSOX1,GSN,DKK1 were all highly expressed in PLC patients,which may be closely related to the occurrence ofPLC.2.QSOX1,GSN,DKK1 have a certain reference value for the diagnosis of PLC.3.The application of binary logistic regression model to establish the best QSOX1,GSN,DKK1 regression equation maybe can improve the efficiency of the diagnosis of PLC,which has important reference value to the diagnosis of PLC.4.The expression level of QSOX1,GSN and DKK1 is not considered to be associated with the prognosis of patients with PLC,and the BCLC stage,CA125 expression,and treatment modalities may be independent prognostic factors for PLC in patients with PLC.
