Objective: By detecting the degeneration degree of rabbit nucleus pulposus through different acupuncture methods,to establish a animal model which should be closer to the clinical testing through imaging,anatomy and enzyme-linked immunosorbent assay.Through vivo injection of Bone-morphogenetic protein-7 in treating rabbit degenerative nucleus pulposus,validating the influencesontype collagen,which could be an reference Ⅱfor further research on factors in treating lumbar intervertebral disc degeneration,still in reference for the clinical treatment of lumbar degenerative,it might provides new methods.Method:Preparing18 adult healthy New Zealand white rabbits,all 4 discs of each rabbit between L2~L6 were selected as research objects,the18 rabbits were divided into 3 groups randomly,both puncture and rotating were performed on the discs in group A,nothing but puncture was performed on the discs in group B,no rotating nor puncture in Group C which was set for blank group,with no special processing.Validating took place respectively in1 week,and 2 weeks,and 4 weeks after the operation to check if it was a operative model of rabbit disc pulpy nucleus degenerative.After the model validation was successful,48 adult healthy New Zealand White Rabbit were prepared,all 4 discs of each rabbit between L2~L6 were selected as research objects,48 rabbits were randomly divided equally into 4 groups and given special model number for each one.BMP-7 was injected within and the around the disc pulpynucleus in the treatment group,muscle note antibiotics to prevent infection for three days postoperatively,with no other processing.Infiltration salineset was injected within and the around the disc pulpy nucleus in the control group,muscle note antibiotics to prevent infection for three days postoperatively,with no other processing.No treatment was performed on the blank group,but muscle note antibiotics were still given to excluded the interference of antibiotics.In addition,anther group was set to contrast with the treatment group in control research,with the same treatment in the treatment group at the same time,yet sedatives were given to reduce the motion.Check respectively in2 weeks,and 4 weeks after the operation,as well as the blank Group,therefore,each group was divided into 4subgroups vertically,there were 3 rabbits in each subgroup which contained 12 research objects.Contrast the test results of each subgroup horizontally and vertically,the test method concluding:(1)Body Weight Changes,(2)mark the disc height by x-ray photography,anatomy observation,detect the type(3)(3)II collagen protein content in disc pulpynucleusby ELISA.Results: Group A and group B had developed a similar degeneration rate based on the outcomes and statistical results,but group A had made a higher success rate,its repeatability was more stable and reliable.There was a horizontally contrast in the treatment situation,the disc height of the treatment group was higher than that in the control group in X-ray performance with statistically significance.There were no obviously differences between the control group and the blank group.Fresher discs,clearer structure and lesser scar tissues were found in the treatment group,while the discs were scartissues filled in the control group and the blank group.No obviously differences were found between the control group and the blank group.More type II collagen protein was found in the treatment group than that in the control group and the blank group by ELISA with statistically significance.Still,no obviously differences were found between the control group and the blank group.A vertical comparison was made inside each group,there was no statistically significance in the treatment group including the change of disc height in X-ray performance,the scarring progress in anatomy observation,the type II collagen protein content in the ELISA.While statistically significance was found in the control group and the blank group through the same comparison.There was no statistically significance between the treatment group and the sedation group.Conclusion: Both puncture and rotating performed on the rabbits’ discs can create a satisfied model of nucleus pulposus of intervertebral disc degeneration,BMP-7 could degenerate the nucleus pulposus effectively by reduce the loss of type Ⅱcollagen which means exact effect in vivo treatment of the rabbit nucleus pulposus of intervertebral disc degeneration.
