Obesity in human and animals is considered to be the result of excessive proliferation and differentiation of preadipocytes.Inhibiting the excessive proliferation and differentiation of the preadipocytes will be beneficial to the human diet,prevent and reduce obesity related diseases.It is reported that the effect of reducing weight and lowering blood sugar is mainly based on the study of the body.This paper uses 3T3-L1 preadipocytes to reveal the effect of plant polysaccharides on the fat cells from the cell level and molecular level.The main contents are as follows.Using gas chromatography to analyze the composition of five kinds of polysaccharides(Green tea polysaccharide,Black tea polysaccharide,Oolong tea polysaccharide,Lycium barbarum polysaccharide,Oat polysaccharide,GTP,BTP,OTP,LBP,OP).We used MTT method to determine the effects of five kinds of polysaccharides on the proliferation of 3T3-L1 preadipocytes.Cell cycle was determined by flow cytometry,using PI staining.The preadipocytes cells were differentiated into adipocytes by the traditional "cocktail" method.Oil red O staining and triglyceride content were measured to analyze the differentiation rate and accumulation of lipid content in adipocytes.The expression of the related adipocyte factors and major adipocyte transcription factors were analyzed by RT-PCR.It was found that tea polysaccharide(TP)was composed of glucose,arabinose,rhamnose,mannose and galactose,and LBP was composed of arabinose,rhamnose,galactose,and glucose,but the main components of oat polysaccharide(OP)was glucose.High concentration of TP had some toxic effects on the cells.On one hand,in a certain concentration range,TP and OP can inhibit the proliferation of 3T3-L1 preadipocytes.At the same time,the number of cells in G1 phase were increased by TP and OP but the number of cells in S phase were decreased.LBP can promote the cell proliferation.On the other hand,TP and OP can inhibit the differentiation of 3T3-L1 preadipocytes,and decrease the content of TG,but LBP can promote the differentiation of 3T3-L1 preadipocytes and increase the content of TG.And GTP,BTP,OTP,OP can up-regulate the expression of ADP and down-regulate SREBP-1c,PPAR?,LPL,FABP4,SCD1,FAS,ACC expression in differentiated adipocytes,but the results of the LBP group was just the opposite.The results showed that LBP promote the proliferation and differentiation of 3T3-L1 preadipocytes.TP and OP were able to inhibit the proliferation and differentiation of 3T3-L1 preadipocytes,and they could affect the cell cycle,regulation of lipid metabolism,which was associated with down-regulating expression of SREBP-1c,PPAR ?,LPL,FABP4,SCD1,FAS,ACC and up-regulating expression of ADP.
