Objective:Melasma is a common skin condition characterized by disorders of melanin pigmentation or accumilation of the face coursing by multifactor effective factors.This experiment is to explore the in vivo and in vitro pharmacocdyniamics of Qing-ban cupsule on the prevcention of and the ablity of anti-oxidation in order to observe the treatment effect of Qing-ban cupsule on Melasma.As to provide scientific basis for clinical application and offer reliable experimental metlhods and theory of Qing-bain calpsule.Methods:(1)formulation research of Qing-ban capsule.(2)Prepare chloasma model mice induced by injection of progesterone in vitro,SOD,MDA,GSH were detected by kit in scrum and liver after drug treatiment;vitro effects on tyrosinase activity after drug treatment;melanoma of skin in mice was obscervced by immuno-histochemistry.(3)B16 marine nelanonmal cells were used in whitening effect assay,incubated with different concentratioins of Qingban Capsules for 72h,and the quinhydrone were selected as positive controls.MTT method,NaOH assay and DOPA oxidization reaction was employed to determine the cell proliferation,melanin synthesis,tyrosinase activity of melanocytes.Results:(1)Qing-ban capsule could significantly improve the SOD,GSH activity and decrease MDA content in a concentration-dependent manner.The high-dose and mid-dose group were higher then conrtal group significantly(p<0.05),which improve the SOD,activity and decrease MDA content.(2)In the control group,there were no melanocytes express in epidermis,barely seen in hair follicles and sebaceous basal cell,and lighter coloring,showing a pale yellow.Qing-ban cupsule could reduce the expression of melanocyte.(3)In vitro experiment showed that the capsule significantly improv restrained the activity of tyrosinase,especially the high concentrations of Qing-ban capsule,the content was statistical significance(P<0.05).(4)Under the condition of different,Qing-ban cupsule can inhibit the proliferatio?tyrosinase activity and Melanogenesis on B-16 Murine Melanoma,which was remarkable than that of contral group(p<0.05).It was found that Qingban Capsules can restrain the proliferation,tyrosinase activity and melanin synthesis of melanocytes in a concentration-dependent manner,the content was statistical significance(P<0.05).Conclusions:(1)Qing-ban capsule can significantly improve the SOD,GSH activity,decreased MDA content;In vitro experiment showed that the capsule significantly improved the activity of tyrosinase,inhibit the fonnation of melanin in skin tissue.It was also found that Qingban Capsules restrain the proliferation,tyrosinase activity and melanin synthesis of melanocytes in a concentration-dependent manner.(2)The mchanism is not a single way in the treatment of melasma of Qing-ban capsule.Qing-ban capsulesuppress of melanocyte proliferation,tyrosinase activity and improve the body's SOD activity,to remove excess oxygen free radicals to treat melasma.(3)Qing-ban capsule shows good anti-oxidation activity and has good treatment on chloasma model mice induced by progesterone.
