Objective: 1. In our study, we investigated the expression of miR-9, miR-21 and miR-501 in serum and plasma of patients with non-small cell lung cancer (NSCLC),and explored whether miR-9, miR-21 and miR-501 could be sensitive biomarkers of miRNAs in peripheral blood of patients with small cell lung cancer.2. To analyze the difference between the expression of miRNA in serum and plasma of patients with non-small cell lung cancer and explore the levels difference of miRNA expression between serum and plasma in patients with non-small cell lung cancer.3. We analyzed the relationship between the expression of miRNAs and clinicopathological parameters in patients with non-small cell lung cancer (NSCLC).And we explored whether the miRNAs might be used to monitor the recurrence and metastasis of non-small cell lung cancer, or evaluated the efficacy of treatment and prognosis. Whether it is a specific biomarker of Xuanwei lung cancer.Methods: 1. Collect the data of the patients who met the inclusion criteria. After the second day of admission, the samples were collected from the fasting venous blood(anticoagulant 10ml, non-anticoagulant 10ml), and the serum and plasma were stored in a clean centrifuge tube Store at -80 ?, and we set healthy for the control group.2. Serum and plasma samples were collected from healthy subjects and non-small cell lung cancer patients. MiR-9, miR-21 and miR-501 were amplified by qRT-PCR. The expression of miR-9, miR-21 and miR-501 was detected by qRT-PCR.The levels of miR-9, miR-21 and miR-501 were compared. The formula for the relative expression of miR-9, miR-21 and miR-501 is: 2-??CT.3. MiR-9, miR-21 and miR-501 were analyzed statistically by group of Xuanwei and non-Xuanwei, early and middle stage, male and female.4. The relationship among the expression levels of miR-9, miR-21 and miR-501 in serum or plasma, the clinicopathological parameters of lung cancer patients and the level of serum tumor markers.5. The sensitivity and specificity of miR-9, miR-21 and miR-501 and serum tumor markers CEA,CA125, CA153, CA199, CA242, CA724, FER, CYFRA,NSE were measured, And we calculated their under the area of the ROC curve.Results: 1. MiR-9, miR-21 of Serum and plasma in healthy people and non-small cell lung cancer patients with high levels of expression, the difference was statistically significant (P <0.05), miR-501 in healthy people and non-small cell lung cancer the difference was not statistically significant (P> 0.05).2. The expression level of serum miR-9 in healthy group was 1.15 ± 0.76 and 3.39 ±3.75 in non-small cell lung cancer group. Compared with healthy group, the expression level of miR-9 in non-small cell lung cancer group was higher than that in healthy group was statistically significant (P <0.05). The expression level of miR-21 in healthy group was 0.87 ± 0.29, 2.3 ± 3.02 in non-small cell lung cancer group.Compared with healthy group, the expression level of miR-21 in non-small cell lungcancer group was higher than that in healthy group (P <0.05). The expression level of miR-501 in healthy group was 1.84 ± 2.04, 2.30 ± 3.02 in non-small cell lung cancer group. Compared with healthy group, the expression level of miR-501 in non-small cell lung cancer group was higher than that in healthy group Meaningful (P> 0.05).The expression level of miR-9 in the healthy group was 1.12 ± 0.70 and that in the non-small cell lung cancer group was 3.41 ± 3.73. Compared with the healthy group,the expression level of miR-9 in the non-small cell lung cancer group was higher than that in the healthy group Meaningful (P <0.05). The expression level of miR-21 in healthy group was 0.84 ± 0.32 and 2.37 ± 3.04 in non-small cell lung cancer group.Compared with healthy group, the expression level of miR-21 in non-small cell lung cancer group was higher than that in healthy group Meaningful (P <0.05). The expression level of miR-501 in the healthy group was 1.84 ± 2.06 and that in the lung cancer group was 3.27 ± 3.60. Compared with the healthy group, the expression level of miR-501 in the non-small cell lung cancer group was higher than that in the healthy group, the difference was not statistically significant (P> 0.05).3. The levels of mir-9, miR-21 and miR-501 in serum and plasma of non-small cell lung cancer were significantly higher than those in non-small cell lung cancer group(P>0.05), and the correlation coefficient was close to 1, P <0.001.4. The relationship between expression levels of miR-9 and miR-21 in peripheral blood circulation and patients' clinicopathological parameters suggesting that the expression of miR-9 and miR-21 was not correlated with whether patients lived in Xuanwei, the expression of miR-9 and miR-21 was not correlated with gender, age,TNM stage and metastasis (P> 0.05). MiR-21 was not associated with pathological type (P> 0.05). There was a correlation between miR-9 and pathologic type (P <0.05).5. The increase of miR-9 level in peripheral blood circulation was related to the increase of NSE and Cyfra21-1 (P <0.05). The level of miR-21 in peripheral blood circulation was related to the increase of FER,Cyfra21-1,NSE and CA153,(P<0.05).6. The specificity and sensitivity of miR-9 and miR-21 in the detection of non-small cell lung cancer were not significantly better than those of tumor markers CEA,CA125, CA153, CA199, CA242, CA724, FER,CYFRA-12, NSE,SCC,where the area under the ROC curve of miR-9 reached 0.75, with general diagnostic.7. The combination of 4 kinds tumor markers (miR-9, CEA,CA724, CYFRA21-1)were high specific and strong sensitive.Conclusion: 1. MiR-9 and miR-21 were highly expressed in peripheral blood circulation of patients with non-small cell lung cancer, this phenomenon suggesting that the expression levels of miR-9 and miR-21 in peripheral blood circulation can be used for the diagnosis of non-small cell lung cancer, miR-9 and miR-21 have the potential to be biomarkers of non-small cell lung cancer.2. Peripheral blood circulation miRNA expression in serum and plasma contrast is no difference.3. Peripheral blood circulation miRNA expression in serum and plasma contrast is no differences. The combination of miR-9 and CEA,CA724, CYFRA21-1 expression levels is more specific and sensitive. And this way can improve the accuracy of non-small cell lung cancer diagnostic and reduce the false positive rate.
