Objective By using the animal experiments to observe the effect of Suhuang Xiezhuo Pill on TGF-?1/p38 MAPK transduction pathway in the renal interstitial fibrosis of UUO model rats.To investigate the mechanism of Suhuang Xiezhuo Pill on anti-renal interstitial fibrosis,and to provide a theoretical basis for the treatment of renal interstitial fibrosis.Methods 70 specific pathogen-free male Wistar rats weighed 180-210 g were randomly selected.The rats were randomly divided into control group,sham operation group,UUO model group,Suhuang Xiezhuo Pill group,and Niaoduqing group.The renal interstitial fibrosis model was made by unilateral ureteral ligation,and the left ureter was ligated to make an UUO animal model.After successful modeling,the control group,sham operation group,and model group were respectively given saline(100g-1ˇml-1ˇday-1,i.g)according to the weight of the rats.The Suhuang Xiezhuo Pill group was given proportionally prepared solution(100g-1ˇml-1ˇday-1,i.g).The time of gavage was fixed in each day.The experiment period was 28 days.The situation of the rats was observed.The varieties of serum creatinine,urea nitrogen and urinary albumin after drug administration were detected on the 14 th day and the 28 th day.The degree of renal interstitial fibrosis was observed by MASSON staining method.The immunohistochemistry was applied to detect the expression of TGF-?1 and p38 MAPK.Results The rats showed no significant differences before the model establishment.On the 14 th day of successful modeling,the serum creatinine,urea nitrogen and urinary albumin contents of the model group were significantly increased than that in control and sham operation group.Comparing with the control group and the sham operation group,the indexes of the drug administration group were increased,but were lower than those in model group.The results of the laboratory examination were better in Suhuang Xiezhuo Pill group than those of the Niaoduqing group.On the 28 th day of successful modeling,the renal interstitial fibrosis in the model group showed an obvious aggravation.The serum creatinine,urea nitrogen and urinary albumin contents of the model group on the 28 th day were higher than those on the 14 th day.There were small changes of indexes in drug administration group.There were no obvious changes of the laboratory examination between the 14 th day and the28 th day.The serum creatinine,urea nitrogen and urinary albumin contents in Suhuang Xiezhuo Pill group were lower than those in Niaoduqing group.The Suhuang Xiezhuo Pill group showed a lightest renal interstitial fibrosis among all the groups.Conclusion The Suhuang Xiezhuo Pill ameliorated the renal interstitial fibrosis of UUO model rats.The serum creatinine,urea nitrogen and urinary albumin contents were decreased in Suhuang Xiezhuo Pill group.The expressions of TGF-?1 and p38 MAPK in renal tissues were reduced by Suhuang Xiezhuo Pill,and the progress of renal interstitial fibrosis was delayed by Suhuang Xiezhuo Pill.
