Objective:HCC is one of the morbidity and mortality of malignant tumors,diagnosis and treatment of hepatocellular carcinoma recent years,China has made great achievements.Because of the large number of people infected with HBV and HCV in China,hepatocellular carcinoma will still be a major disease that threatens the health of our people.Therefore,we will explore the genes related to the development and progression of hepatocellular carcinoma.The aim of this study is to explain the mechanism of hintl mediate miRNA-455-5p/CD47 in hepatocellular carcinoma,it is expected to provide a new Hint pathway to regulate the invasion and metastasis of hepatocellular carcinoma cells.Methods:1.The Hintl gene was knocked out with siRNA in HepG2 cells,and the knockout ability was confirmed by qPCR.The invasion and metastasis ability of HepG2 cells knocked out hintl was determined by the scratch test and Transwell assay.2.The expression of Hintl was detected by gene chip technique.And the expression of CD47 in the HepG2 cells knocked out of hintl was determined by qPCR.4.The bioinformatics software used was TargetScanF6.2(www.targetscan.org)test and verify that miRNA-455-5p is target protein of CD47,5.qPCR was used to determine the expression of miRNA-455-5p in HepG2 cells knocked out hintl;6.miRNA-455-5p was overexpressed with agomir and verified by qPCR.The expression of CD47 protein was detected by western blotting in HepG2 cells knocked out of hintl and overexpressed miRNA-455-5p.The invasion and metastasis were detected by scratch test and Transwell assay.Results:1.In the HepG2 cells knocked out of the hint1 gene,qPCR showed a decrease in hintl expression,and the results of the scratch test and Transwell showed that the cell invasion and metastasis were increased.2.The results showed that the expression of CD47 was the most obvious in the expression of CD47.3.In the HepG2 cells knocked out hintl gene,qPCR showed overexpression of CD47.4,TargetScan 6.2 showed that the target protein of mRNA was CD47.The expression of miRNA-455-5p in HepG2 cells of hintl was decreased.QPCR showed that the expression of miRNA-455-5p was increased after the miRNA-455-5p was overexpressed by agomir.7.In the HepG2 cells knocked out hintl,qPCR showed that CD47 expression was down-regulated,scratching,Transwell showed HepG2 cells invasion,transfer capacity is suppressioned.Conclusion:1 Knockdown of hintl HepG2 cells increased the ability of invasion and metastasis.;2 hintl inhibits the:invasion and metastasis of HepG2 cells by inhibiting CD47 in hepatocellular carcinoma cells;3 miRNA-455-5p inhibits the invasion and metastasis of HepG2 cells by inhibiting CD47;4 HINTl mediates miR-455/CD47 signaling pathway,inhibits hepatocellular carcinoma migration and invasion?...
