Expression microarray technology produce a large number of gene chip and mass biological information. It is the challenges and goals for biological workers that using the microarray data from numerous laboratories and reading the biological meaning of the complex gene expression data.The occurrence and development of HCC is a multi-stage, multi-step and polygenic complex process. The traditional research methods of the molecular mechanism of hepatocellular carcinoma are mainly single gene, and the results have some limitations. The research of gene chip can display high-throughput gene changes, which have a greater advantage to the molecular mechanism of the pathogenesis of hepatocellular carcinoma. The objective of this study is analyzing hepatocellular carcinoma microarray from gene chip database by bioinformatics, screening the differentially expressed genes, and exploring the molecular mechanism of hepatocellular carcinoma.Methods:Gene chip data GSE60502 is downloaded from the GEO database. The microarray data are analyzed by Genespring software,which screen the differentially expressed genes.Using online software DAVID analyze biological function and Pathway enrichment of significantly differentially expressed genes.Results:1. A total of 1067 significantly differentially expressed genes is confirmed,including 626 up regulation and 450 down regulatiaon.2. GO functional annotation results: the differentially expressed genes of up regulation mainly relate to the cell cycle phase M, mitosis, purine nucleotide binding, etc. the differentially expressed genes of down regulation mainly relate to microsome, acute inflammation, protein degradation, complement activation, positive regulation of immune response, oxidoreductase activities, etc. 3. The pathway enrichment results: the differentially expressed genes of up regulation mainly focus on the cell cycle, reduction division of oocytes, Progesterone- mediated oocyte maturation, P53 signaling pathway, cancer; the differentially expressed genes of down regulation mainly focus on: Complement and coagulation cascades,drug metabolism, retinol metabolism, heterosexual biomass of the cytochrome P450 metabolic, linoleic acid metabolism.Conclusion:1.MAPK signaling pathway may be one of the main molecular pathogenesis of HCC;2.The upregulation of p53 signaling pathway has certain protective effect to the occurrence of HCC;3.The downregulation of CYP450 has some degree of significance for the clinical use of drugs and cancer prevention.
