Background:In clinical practice,the gallbladder cancer is one of the more common malignant tumors in hunman's biliary tract system.In recent years,the morbidity of gallbladder cancer trends to raise year by year.Since gallbladder cancer lacks of specific clinical symptoms in the early time and lower accuracy rate and specificity to diagnose by imageological examination.It is difficult for most of the patients with gallbladder cancers to diagnose and treat in a earlier time.Most of them are often diagnosed in the advanced stage and lose the opportunities to make a radical surgical operation resection.On the other hand,the patients with gallbladder cancer have an extremely poor prognosis.For these reasons,the patients with gallbladder cancer have an extremely lower five-year survival rate and a quite high mortality.So it is necessary to find another method to cure gallbladder cancer patients by making clear of its pathogenesis.The methods to cure and diagnose malignant tumor at the genic level are more and more grown at present.In recent years many researchers believe Eukaryotic translation initiation factor 5A2 is a dubious carcinogenic gene.The DNA mRNA and protein content of it can be find in many kinds of human malignant tumors.We find that it can make malignant tumor grow more fast and disordered by make more researchs.But the research reports in gallbladder cancer can only be find in our teams works.Objectives:1.To study the gene expression quantities of Eukaryotic translation initiation factor 5A2 in the gallbladder cancer.2.To make sure the relationship between the gene expression quantities of EIF-5A2 and the differentiation and migration of gallbladder cancer.3.To explore the relationships between the gene expression quantities of EIF-5A2 and patient's clinical data.Methods:Collect the patients who received the laparoscopic cholecystectomy and gallbladder radical surgery in Second Affiliated Hospital of Kunming Medical University in January 2013-May 2016.Search the pathological specimen of these patients and use the Reverse transcription-polymerase chain reaction to get the value of 2-??t which can represent the gene expression quantities of EIF-5A2.Study the relationships between the gene expression quantities of EIF-5A2 and the differentiation and migration of gallbladder cancers,patient's clinical data by multiple linear regression analysis,t test and analysis of variance test.Results:1.The 2-??t of chronic cholecystitis and atypical hyperplasia is 2.4621±0.77748,and the gallbladder cancer's is 8.9641±11.03640.It is statistically significant between them(t=-2.978,P=0.006<0.05).2.The 2-??t of poorly differentiated adenocarcinoma,moderately differentiated adenocarcinoma,high differentiated adenocarcinoma and other genre tumor are 16.9134±15.20265,4.3392±2.00329,2.9595±1.04541,5.5447±3.07981.It is statistically significant among them(F=3.645,P=0.028<0.05).Make Least Significant Difference.The compare between high differentiated adenocarcinoma and poorly differentiated adenocarcinoma is statistically significant(P=0.015<0.05).It can also be seen between poorly differentiated adenocarcinoma and moderately differentiated adenocarcinoma(P=0.015<0.05).On the contrary,the value between moderately differentiated adenocarcinoma and high differentiated adenocarcinoma isn't statistically significant(P=0.809>0.05).Even more,the value of other genre tumor is only lower than poorly differentiated adenocarcinoma(P=0.042<0.05).3.Compare the value of 2-??t among poorly differentiated adenocarcinoma,poorly differentiated adenocarcinoma with lymphatic metastasis,moderately differentiated adenocarcinoma and moderately differentiated adenocarcinoma with lymphatic metastasis,2.8876±0.61913,6.2746±1.27726,6.6753±3.78602,23.7388±16.36024.It is statistically significant between moderately differentiated adenocarcinoma and moderately differentiated adenocarcinoma with lymphatic metastasis(P=0.005<0.05),and it isn't statistically significant between poorly differentiated adenocarcinoma,poorly differentiated adenocarcinoma with lymphatic metastasis(P=0.079>0.05).4.The distinction of sex and the numbers of pregnancy can't influence the value of 2-??t(P=0.5090>0.05),(P=0.839>0.05).5.The gene expression quantities of EIF-5A2 is significantly correlated with migration conditions(t=3.160 P=0.005<0.05)but not with the numerical value of age of patients(r=0.247 t=0.074>0.05).6.The test positive rate between traditional tumor marker and EIF-5A2 test are not statistically significant(P=0.528>0.05).7.The gene expression quantities of EIF-5A2 is not significantly correlated with calculus exists or not,is not significantly correlated with diameter of the tumors(F=0.526 P=0.598>0.05).Conclusions:1.EIF-5A2 can expression in all kinds of gallbladder tissue,but is higher in the gallbladder cancer tissue.2.The gene expression quantities of EIF-5A2 in the gallbladder cancer will increase with the tumor stage growths.And There is high relevance between gene expression quantities of EIF-5A2 and the migration conditions in human gallbladder cancers.3.The gene expression quantities of EIF-5A2 are not significantly correlated with the numerical value of age,distinction of sex,the numbers of pregnancy,diameters of the tumors and calculus exists or not.
