The Effect Of Injecting Different Doses Of Dopamine D1-like Receptor Antagonist On Pakinson’s Motor Behavior In Rats

Abstract/Summary:
In this research,we injection 6-hydroxydopamine(6-OHDA)into the medial forebrain bundle to establish Parkinson’s Disease(PD)model in rats.Using some indexes to simulate PD patients’ motor and non-motor behaviors,and then have an evaluation about them.The indexes consists of numbers of touching the wall,the percentage of damaged forelimb touching the wall,cacation number in Cylinder Test and central area time,rest time,vertical and horizontal score,modified number and cacation number in Open Field Test.Observing changes of PD rats’ behaviors after intraperitoneal injecting dopamine(DA)D1 receptor antagonist SCH-23390,then make preparations for later electrophysiological experiments.Aims:1.To explore the effect on motor and non-motor behavior of PD rats after injecting different concentration of SCH-23390;2.To find a suitable concentration of SCH-23390 for later electrophysiological experiments;3.To explore if SCH-23390 have a therapeutic effect on PD rats.Results:The results of Cylinder test:Efficacy of time measurement results: injecting 0.02 mg/Kg body weight of SCH-23390,in 15~20 min period,the numbers of touching the wall reduced,in 25 ~30 min and 35~40 min period,the numbers of touching the wall greatly reduce,so we choose 25~30 min after injection to go on behavioral science experiments.After injection 0.01 mg/Kg SCH-23390,number of touch the wall in normal rats and PD rats are decreased significantly,with the concentration increased,the wall number decreased sharply,there is a significant difference in two groups.When the concentration reach 0.05 mg/Kg,there is no difference between the two groups.After injecting SCH-23390,there is no difference in PD rats and normal rats on percentage of damaged forelimb touching the wall,but PD rats have a higher percentage than normal rats.As for the cacation number within the two groups,no difference emergedIV before and after concentration.Injecting kinds of SCH-23390 for 10 days and collecting these indexes again,there is no difference compared with 10 days before.The results of Open Filed Test:Normal rats injected with concentration of 0.005 mg/Kg body weight after SCH-23390,significant growth fringe area rest time,the rest of the various indicators and dosing before had no difference;Injection concentration of 0.01 mg/Kg body weight after SCH-23390,the central time significant growth,level score and vertical scores were significantly reduced,modified number and grain number defecate no difference.With the increase of concentration of modification times and defecation particle number and the medication before had no difference,the other indexes had significant difference.PD rats injected with concentration of 0.005 mg/Kg body weight after SCH-23390,significant growth fringe area rest time,the rest of the various indicators and dosing before had no difference;Injection concentration of 0.01 mg/Kg body weight after SCH-23390,the central time significant growth,across the g score and the number of defecation particles decreases,the rest of the indicators and dosing before no difference;Injection concentration of 0.02 mg/Kg body weight after SCH-23390,vertical score to reduce;Before the number with the increase of concentration of modifier,and the drug no difference,the other indexes have difference.Compared with normal rats and PD group rats found that,after injection of different concentration SCH-23390 central time between the two groups of rats were significant difference;When the concentration of injection of 0.005,0.01,0.02 mg/Kg body weight,the fringe area to have a rest time between two groups of rats and vertical score had significant difference,but when the concentration of 0.05 mg/Kg body weight no difference between the two groups;When injection of saline,horizontal score between two groups of rats and defecation particle count has significant difference,injection SCH-23390 concentration was 0.005 and 0.01mg/Kg body weight,the score levels between the two groups and defecation particle number have difference,injection concentration of 0.02,0.02 mg/Kg body weight,no difference between the two groups.Injecting kinds of SCH-23390 for 10 days andcollecting these indexes again in Open Field Test,there is no difference compared with 10 days before.Conclusion:SCH-23390 makes PD rats and normal rats have lower sports ability and PD rats characterized anxiety and tension;Compared with SCH-23390’s influence on rats’ motor and non-motor behavior,we choose 0.02 mg/Kg body weight as later electrophysiological experiment concentration.SCH-23390 has no treatment effect on motor and non-motor behavior.
