Objective:Sangjuyin Decoction is a famous prescription in Wenbing Tiaobian,which is consist of mulberry leaves,chrysanthemum,forsythia suspensa,peppermin,licoric,reed root,almonds and platycodon and commonly used to disperse wind,clear away heat and diffuse the lung,suppress cough.In present,it is widely used in the treatment of exogenous cough,acute bronchitis,upper respiratory,pneumonia and other respiratory disease in clinical practice.Modern pharmacological studies have shown that Sangjuyin Decoction has significant anti-inflammatory,antimicrobial,anti-oxidation and protect mitochondria,relieve acute lung injury,antipyretic and sweating,inhibit effect on intestine peristalsis and improve immune function,but the mechanism is unclear.In this paper,as a guidance for the Chinese medicine theory,the use of modern technologies,from the whole animal,using the mouse model of cough,study the Sangjuyin Decoction and its ingredients anti-cough mechanism of preliminary study and provide scientific basis for clinical application and further development of Sangjuyin Decoction.Methods:The model of cough mice were made using capsaicin spray.Mice were randomly divided into control group,model group,high dose,middle dose and low dose of Sangjuyin Decoction and its ingredients.Combining our previous study with the number of mice cough,pathological examination,amount of SP release,we determine the successful model.The Sangjuyin Decoction and its ingredients or saline were adminnistraed for 5 days after being made successfully.The trachea and bronchus tissue,trigeminal ganglion were also collected.The pathological change of trachea and bronchus tissue was observed under light microscope after HE staining.RT-PCR and Western Blot method was undertaken to test the expression of TRPV1 in trachea and bronchus tissue.Meanwhile,the expression level of SP were determined by ELISA.Results:The mice of model group exist in active,irritability and airway inflammation.Meanwhile,the times of cough,the release of SP and TRPV1 receptor relative expression in trachea and bronchi of model group mice were higher than that of control group in saline,indicated that the cough model was successful.After treatment of Sangjuyin Decoction,compared with the model group,we found significant declines in the times of cough,the release of SP and TRPV1 receptor relative expression in trachea and bronchi of high and middle groups.The effect was not obvious in the lower dosage group.After treatment of Sangjuyin Decoction ingredients,the above mentioned indexes seems lower than before,but the results showed no statistically significant.These indexes in model groups have a significant distinction than those of normal groups.Conclusion:1.The mice cough model was successfully established in this experiment by capsaicin spray.2.The cough symptoms,the relative expression of TRPV1 receptor and release of SP in trachea and bronchi of mice were reduced in high and medium dosage groups.3.There was no significant change in the number of coughs in dismantle square groups,the relative expression of TRPV1mRNA seems lower than before in some groups,but there were no significant or substantive changes in the expression of TRPV1 protein.4.The downgrading of the TRPV1 receptor expression may be one of the biological mechanisms for treating coughs.
