ObjectiveTo investigate the changes of the expression of TRPV1 and substance P in the tongue and trigeminal ganglion of ovariectomized(OVX)rats and the effects of estrogen and remifemin treatment on these changes..MethodsThirty-two female adult Sprague-Dawley rats were randomly divided into four groups: sham operation group(SHAM),ovariectomy group(OVX),estradiol tretment group(E2),and remifemin treatment group(i CR).After two weeks' recovery,the rats were treated with saline,estradiol or remifemin by gastric perfusion for four weeks.Immunostaining was used to detect the expression of transient receptor potential vanilloid 1(TRPV1)and substance P(SP)in the dorsal lingual epithelium and trigeminal ganglion of rats in each group.Results1.The expression of TRPV1 in the dorsal lingual epithelium of OVX group was increased compared with that of SHAM group(P<0.05).E2 and i CR groups were decreased compared with that of OVX group(P<0.05),and had no difference with that of SHAM group(P>0.05).2.The expression of SP in the dorsal lingual epithelium of OVX was increased compared with that of SHAM group(P<0.05).E2 and i CR groups were decreased compared with that of OVX group(P<0.05),but they were increased compared with that of SHAM group(P<0.05).3.The expression of TRPV1 in the trigeminal ganglion of OVX group was i n c r e as e d c o m par ed w it h t h at of SHAM gr o u p(P < 0.0 5).E 2 a n d i C R g r o u ps decreased compared with that of OVX group(P<0.05),and had no difference with that of SHAM group(P>0.05).4.The expression of SP in the trigeminal ganglion of OVX group was i n c r e as e d c o m par ed w it h t h at of SHAM gr o u p(P < 0.0 5).E 2 a n d i C R g r o u ps we r e decreased compared with that of OVX group(P<0.05),and had no difference with that of SHAM group(P>0.05).Conclusions1.Estrogen decline might be the primary reason for the increase of SP and TRPV1 in dorsal lingual epithelium and trigeminal ganglion of ovariectomized rats.These changes might be crucial in the pathogenesis of paresthesia of tongue in perimenopausal women.2.Estrogen and remifemin decreased the expression of TRPV1 and SP in dorsal lingual epithelium and trigeminal ganglion of ovariectomized rats,which indicated that estrogen and remifemin might play an important role in maintaining nomal sensory function of tongue by regulating the expression of TRPV1 and SP.
