The Clinical Study Of The Relationship Between The Cystathionine-?-Lyase And Breast Cancer

Objective: To test the correlation of cystathionine-?-lyase(CSE)and the clinical features of breast cancer and the relevence of STAT3 and CSE expression in breast cancer tissues.Method: All 72 breast cancer tumor and adjacent non-tumor samples from patients during Jun.2016 to Dec.2016 were recruited from the first affiliated hospital of Zhengzhou University,in Zhengzhou,China.21 breast cancer tumor and 21 adjacent non-tumor samples were analyzed the expression of CSE by immunohistochemistry and 15 breast cancer tumor and 15 adjacent non-tumor samples were detected the expression of CSE and STAT3 by qRT-PCR and western blot.Results:(1)Immunohistochemistry results showed that the positive expression rate of CSE was 71.4%(15/21)and 28.6%(6/21)in breast cancer tissues and adjacent normal breast tissues respectively,which exists significant difference(P<0.05).The expression of CSE has no significant correlation with patient age,tumor size,the expression of ER and P53(P>0.05),but CSE expression is closely related to PR expression,histological grades and lymph node metastasis(P<0.05).(2)qRT-PCR and Western Blot results showed that the positive expression rate of CSE was 53.3%(8/15)in breast cancer tissues.The expression of CSE has no significant correlation with patient age,tumor size,histological grade,ER,PR,P53 and HER2 expression(P>0.05),and but CSE expression is closely related to lymph node metastasis(P<0.05).(3)qRT-PCR and Western Blot results showed that both STAT3 total mRNA levels and STAT3 total protein levels in 15 breast cancer tissues were significantly increased comared with adjacent normal breast tissue.Moreover,STAT3 expression is positively related to the expression of CSE.Conclusion:(1)The expression of CSE in breast cancer is obviously higher than that in the adjacent normal breast tissues.The expression of CSE is closely related to histological grades and lymph node metastasis.CSE may involve the occurrence,development and metastasis of breast cancer.(2)The expression of CSE in breast cancer was positively correlated with STAT3.CSE could promote process of breast cancer in association with the STAT3 signaling pathway.The data may provide the basis for CSE to be a tumor marker and target therapy of breast cancer.
