ObjectiveTo study the effect of Guizhi FulingPills andGui zhi fu ling pill mixed with AG490 liquid of Drugto viability,and expression of associated protein STAT3 and P-STA T3,CyclinD1 and P53 of human breast cancer MCF-7 cell by experiments in vitro,in order to provide a theoretical foundation forGui zhi fu ling pill.MethodTo detect inhibiting growth of human breast cancer MCF-7 Cells for Gui zhi Fu li ng Pills and its mixed with AG490 liquid of Drug of different dose by MTT;To ob serve influence of Gui zhi Fu ling Pills and its mixed with AG490 liquid of Drug t o MCF-7 cells bymicroscope;Detect the cells cycle associated protein STAT3 and PSTAT3,CyclinD1 and P53 by flow cytometry? Result 1 Compared with the negativecontrol group,the cells results of OD of Gui zhi Fu lin g Pills and its mixed with AG490 liquid of Drugeffecting MCF-7cells for 48 h is dec lineand Cell inhibition effect change with dose.2 Results from microscope show that after Gui zhi Fu ling Pills and its mixed with AG490 liquid of Drug effecting MCF-7cells for 48 h,MCF-7 cells happen to morphol ogical changes,Gui zhi Fu ling Pills and its mixed with AG490 liquid of Drug of middle dose group is most obvious.3 Results of cycle associated protein from flow cytometry show that,comparing wit h blank control group,after Gui zhi Fu ling Pills effecting MCF-7cells for 48 h,the e xpression of cell cycle protein P-STAT3 had inordinately decline(P<0.05),protein Cy clinD1 high dose group had a most obvious decline(P<0.05),low dose group hadn't a obvious decline(P>0.05).The expression of protein P53 high dose group had a raise(P<0.05),low dose group hadn't a obvious raise.comparing with blank control grou p,the level of expression of protein STAT3 had not a statistical significance in the difference concentration group(P>0.05).4 After 48 hs of using the Gui zhi Fu ling Pills and its mixed with AG490 liquid of Drug,the cell cycle protein P-STAT3 had a obviousdecline,Compared with the negativ e group,CyclinD1 protein had a declineof high dose group(P<0.05),protein P53 had a raise of difference dose group(P<0.05),STAT3 difference dose group had not a stat istical significance(P>0.05).Conclusion 1 Gui zhi Fu ling Pills and its mixed with AG490 liquid of Drug all can inhibit pro liferation activity of human breast cancer MCF-7 Cells,make their morphologies cha nge and inhibition rate toMCF-7 cells change with Different doses at the same time.2 Gui zhi Fuling Pills and its mixed with AG490 liquid of Drug can control prolifer ation activity of MCF-7 Cellsby decline expression of protein P-STAT3.Guizhi Fulin g Pills and its mixed with AG490 liquid of Drug can block cells at DNA synthesis stage by decline expression of the cycle protein CyclinD1 and raise expression of pr otein P53.3 The signal paths EGFR and STAT3 aren't only important paths inhibiting proliferat ion activity of MCF-7 Cells,Gui zhi Fu ling pills can inhibiting proliferation activit y of MCF-7 cells by other signal paths.
