Objective: To observe the effects of high glucose on the proliferation of rat mesangial cells,to investigate the effect of different concentrations of artesunate on expression of proliferation and apoptosis of rat mesangial HBZY-1 cells cultured in high glucose and inflammatory factor TNF-alpha,IL-6.Methods: rat mesangial cells were divided into low glucose control group,high glucose model group,micro dose artesunate group,low dose artesunate group,medium dose artesunate group,high dose artesunate group,enalapril drug control group.Morphological changes of glomerular mesangial cells were observed under inverted microscope.The cells were observed 24 h,48h,72 h proliferation by MTT method;detect the rate of apoptosis by flow cytometry;the amount of tumor necrosis factor alpha cells ?(TNF-?)and interleukin 6(IL-6)in the supernatantwere detected by ELISA test method.Results:(1)About 12 h,artesunate cells were adherent,they were stretching into spindle shaped or irregular shape in 24 h,they were into the logarithm growth period in 48 h,and they can be fusion in 3-4 days.The nucleus was oval or round.After adding medicine,degree of the cell fusion of each group was different.MMT results showed: the inhibition rate of the low glucose control group cells was significantly higher than that of high group by P<0.01;the inhibition rate of different concentrations of artesunate was higher than that of high group by P<0.05,and they were time dependence and dose dependence.(3)Artesunate significantly induced the apoptosis of HBZY-1 cells(P < 0.05)and showed certain dose dependence.(4)ELISA results showed:compared with the low glucose group,the content of TNF-? and IL-6 in cell culture supernatant of high glucose group was significantly higher by P < 0.01;compared with the high glucose group,the content of each group in the supernatant of artesunate TNFalpha and IL-6 were decreased by P < 0.05,and the high concentration of artesunate group was more obvious than that of enalapril group by P < 0.01.Conclusion: high glucose surroundings can promote the proliferation of rat mesangial cells and can lead to inflammatory reaction;the proliferation of high glucose leaded rat mesangial cells can be inhibited by artesunate,they can be a dose and time dependent manner.artesunate can induce the expression of apoptosis in rat mesangial cells and down-regulation of inflammatory factor TNF-alpha,IL-6.It is concluded that artesunate can be used as an effective drug treatment,prevention and treatment of diabetic nephropathy.It is concluded that artesunate can be used as an effective drug treatment,prevention and treatment for diabetic nephropathy,and alleviate the damage of tissues and organs in high glucose environment.
