Objective: The incidence of ischemic stroke increases year by year,which poses a serious threat to human health.In the treatment of ischemic stroke there is a problem that has been troubling us,that is,cerebral ischemia and reperfusion injury.The study found that limb ischemic postconditioning,can activate the body's protective pathway and antioxidant factors,so that cerebral ischemia-reperfusion injury has been effectively improved.The Nrf2 / ARE pathway plays a very important role in the human environment,especially its antioxidant stress.Then the Nrf2 / ARE pathway in limb ischemic postconditioning can be activated,play its role in brain protection? There are no reports yet.In this study,we investigated the effect of Nrf2 and SOD1 protein on the expression of Nrf2 / ARE pathway by activating the post-ischemic postconditioning.Methods: The rats were randomly divided into three groups: sham operation group(S group),cerebral ischemia reperfusion group(I / R group)and limb ischemic postconditioning group(LpostC group).The neurobehavioral scores were scored 24 hours after the model was made;The infarct volume was measured by TTC method;The histopathological changes of the brain tissue were observed by HE staining;The expression of Nrf2 and SOD1 protein was detected by Western blotting.Results: The neurological score showed that the S group was 0 min,no nerve defect,I / R group was 2.98 ± 0.41,LpostC group was 1.87 ± 0.36,LpostC group had better neurological function than I / R group(P <0.05).Cerebral infarct volume measurements showed that the S group was normal brain tissue,I / R group was 35.24 ± 2.18%,LpostC group was 29.73 ± 1.46%,The infarct volume of LpostC group was lower than that of I / R group(P <0.05).HE staining showed no pathological changes in brain tissue of group S;I / R group showed morphological changes in acute ischemic lesions;ischemic lesions in LpostC group were significantly lower than those in I / R group(P <0.05).Western blotting showed that the expression of Nrf2 and SOD1 was extremely low in group S;the expression of Nrf2 and SOD1 in I / R group increased;the expression of Nrf2 and SOD1 in LpostC group was the highest(P <0.05).Conclusions: 1.Postoperative ischemic postconditioning can reduce the cerebral infarct volume and improve the neurological deficit in rats with cerebral ischemia-reperfusion injury.2.limb ischemic postconditioning can make Nrf2 and SOD1 protein expression increased significantly,so that Nrf2 / ARE signaling pathway activate and play the role of antioxidant stress.This suggests that the Nrf2 / ARE signaling pathway may be one of the effective pathways of limb ischemic postconditioning to reduce cerebral ischemia-reperfusion injury in rats.
