Objective:At present,the main means of clinical treatment of premature ovarian failure is hormone replacement therapy,but,a number of clinical studies have shown that hormone therapy will increase endometrial cancer,invasive breast cancer,coronary heart disease,stroke and a series of disease risk.Over the years of studies on relationship between immune system and reproductive function show that reproductive function is regulated by the immune system from birth to after birth.Chitosan Oligosaccharide(COS),one of the basic amino oligosaccharides,which is the only large amounts of natural sugars present.And it has been reported to have an enhanced effect on immune function,a positive effect on mammalian embryonic development,as while as the positive effects on uterine and ovarian function.This study is to observe whether COS has some protective and salvage effects on ovarian senescence and the correlation between this effect and immune function through regular and continuous gavage.Methods:This experiment was divided into two parts:(1)The role of COS in the prevention of ovarian aging(prevention groups): Cyclophosphamide / Busulfan(CY/BUS)(Prevention-low,100 mg/kg.d),medium-dose(200mg/kg.d)and high-dose(300mg/kg.d).(2)The role of COS in the treatment of ovarian aging(treatment groups): Giving intragastric administration of low dose(Treatment-low,100 mg/kg.d),middle dose(Treatment-mid 200 mg/kg.d)and high dose(Treatment-high,300 mg/kg.d)to the pathological ovarian aging models induced by CY/BUS desperately.Pure COS group(200COS,200mg/kg.d)was given intragastric administration COS to normal mice.At the end of the administration,the relevant relationship between ovarian aging conditions and the immunological functions as well as the ovarian functions were detected by measuring the ovarian index and the number of follicles,the content of estrogen and progesterone,the expression of MVH / Fragilis and ?-galactose,spleen index,peritoneal macrophage phagocytosis and immunological factors.Results:(1)In the prevention groups,the thymus index,spleen index and phagocytosis of peritoneal macrophages were increased by COS gavages,as while as the expression of IL-2 and TNF-?.However,and the expression lever of IL-4 was decreased.In the treatment groups showed the same trend.(2)In the prevention groups,the indexes of ovarian function,such as ovarian index and follicle count,estrogen and progesterone content,and the expression of MVH / Fragilis were significantly increased after intragastric administration The expression of p16,p21,p53 and the expression of ?-galactosidase in the ovary were decreased after intragastric administration.In the treatment group,the above indicators showed the same trend.(3)The relationship between ovarian function,ovarian senescence and immune function was analyzed.The correlation analysis showed that MVH / Fragilis mRNAwas consistent with IL-2 / TNF-? mRNA.The expression of p16 / p21 / p53 mRNA consistent with the trend of IL-4 mRNA and negative correlation with MVH / Fragilis mRNAand IL-2 / TNF-? mRNA.Conclusions:(1)COS maintains its function by activating the normal internal morphology of thymus and spleen,activating macrophages,improving the activity of T lymphocytes and B lymphocytes,and balancing the cellular immune and humoral immune mechanisms to maintain the immune function of female mice.(2)COS may play a role in improving immune function in the protection and rescuing of female mice with ovarian function damage and aging.
