Objective? Chronic pulpitis are common problems and frequently-occurring diseases in endodontics. Carious infection is by far the most common etiology for pulpal inflammation and necrosis. As the carious infection progresses from the tooth surface toward the pulp-dentin complex, both the innate and adaptive immunities are activated.Pulpitis has been associated with abundant inflammatory cells, dilated blood vessels,and thickening nerve fibers histopathologically with or without severe pain clinically.IP08 belongs to the importins ? family, nuclear proteins involved in the nucleolar transport. The nucleocytoplasmic shuttling of large molecules is a highly regulated process mediated by specific nuclearimport and export systems. Recent studies have demonstrated that IP08 can regulate nucleolar transport through NF-KB and Smads.IP08 Plays an important role in the process of nucleolar transport. The aim of this study was to investigate the expression of IP08 in chronic pulpitis caused by carious and the mouse first lower molar development.Methods: Its content are divided into two parts.Part1: The purpose of this study was to quantify the expression of IP08 in pulp tissue with clinically diagnosed chronic pulpitis. Healthy pulps acted as controls. 30 healthy controls, 30 chronic pulpitis caused by deep carious human dental pulps were involved in the study. The protein expression and mRNA level of IP08 were detected by immunohistochemistry?immunofluorescence?Western blot and quantitative Real-time PCR,respectively.Part2: The purpose of this study was to investigate IP08 expression in the developing mouse first lower molar. We aim to investigate the spatial-temporal expression patterns of IP08 at different stages of mouse first lower molar development by immunohistochemistry and immunofluorescence.Results:The resulu of part 1: In healthy samples, immunohistochemical and immunofluorescence staining showed positive IP08 expression only in odontoblasts with cytoplasm staining. Under inflammatory conditions, IPO8 staining began to occur innerve fiber tissues and dilated blood vessels. Compared with healthy samples, IP08 expressions at both protein and mRNA levels increased significantly in chronic pulpitis samples.The resulu of part 2: we found that the spatial-temporal expression patterns of IP08 at different stages of mouse molar.Conclusions:1?This study demonstrated that the IP08 may be participate in the formation of dentin and the dental pulp inflammation.2?The results suggest that IP08 may play an important role during the mouse first lower molar development and involved in the formation of primary dentin.
