Diabetes prevention and control is the focus onmedical research and development.In order to explore the hypoglycemic effect and mechanism of Echinops latifolifius Tausch polysaccharide B on experimental type II diabetic rats model,150 male SD rats(weight 200-220g)were provided,fed with high calorie and high sugar diet and injected with streptozotocin at low doses into the rat peritoneal.then,the 150 rat model of type ?diabetes were successfully established.And the abovel50 rats were randomly divided into 7 groups,including diabetic model group,three Echinops latifolifius Tausch polysaccharide B groups at high,middleand low dose,,metformin group,xiaoke pill group and blank control group.During the 49 days of the experience,the weight,feed intake,water intake and the biochemical parameters related to glucose and lipid metabolism,liver and pancreatic histology were detected.The results wereshowed as follows:In Echinops latifolifius Tausch polysaccharide B at high,middle,low three dose groups,the rat weight,muscle,liver glycogen and INS were significantly increased(P<0.01 or P<0.05);In Echinops latifolifius Tausch polysaccharide B at high,middle two dose groups,the fasting blood glucose,glucose tolerance,PG,Ghb,LD,FFA and TNF-awere statistically significant(P<0.01 or P<0.05);In Echinops latifolifius Tausch polysaccharide B at high,middle,low three dose groups,the TC,TG and LDL were significantly decreased(P<0.01 or P<0.05);and HDL only in Echinops latifolifius Tausch polysaccharide B at high dose groupwas statistically significant(P<0.01).In Echinops latifolifius Tausch polysaccharide B at high,high,middle,low three dose groups,hepatocytes showed varying degrees of mild diffuse steatosis,partial shrinkage and different sizes of lipid droplets;In Metformin groupand Xiaoke pill group,the size of the liver cells can be seen within the vacuolar degeneration,nuclear atrophy,swelling,irregular arrangement of sinusoids.In Echinops latifolifius Tausch polymiddle,low three dose groups,the food intake,water intake and ISI were not statistically significant(P>0.05).In Echinops latifolifius Tausch polysaccharide B atsaccharide B at high,middle,low three dose groups,pancreatic acinar cells significantly reduced,?,? cells decreased,atrophy,disorder,acinar secretions decreased.In conclusion:(1)Echinops latifolifius Tausch polysaccharide B can cause the weight loss of type? diabetes model rats slowed down.reduce blood glucose,improve glucose tolerance,under the curve area and glycosylated hemoglobin,increased muscle and liver glycogen content.(2)Echinops latifolifius Tausch polysaccharide B can reduce the total cholesterol,triglycerides,low density lipoprotein,tumor necrosis factor-a,elevated high-density lipoprotein,improve lipid metabolism of type 2 diabetic rats.(3)Echinops latifolifius Tausch polysaccharide B can increase the insulin,reduce glucagon,insulin resistance index,but little effect on insulin sensitivity index,improve islet function of type 2 diabetic rats.(4)Echinops latifolifius Tausch polysaccharide B can upregulate the liver,skeletal muscle,pancreatic insulin receptor,insulin receptor substrate-1,insulin receptor substrate-2 level,improve insulin resistance of type 2 diabetic rats.(5)Echinops latifolifius Tausch polysaccharide B can improve the injury of liver and pancreas tissue in type ? diabetic rats.
